经鼻枸橼酸铁铵处理对小鼠嗅球突触囊泡周期通路的影响
Impact of transnasal administration of ferric ammonium citrate on the synaptic vesicle cycle pathway in the olfactory bulb of mice

Abstract
Objective To investigate the impact of transnasal administration of ferric ammonium citrate(FAC)on the sy-naptic vesicle cycle pathway in olfactory bulb(OB)tissue,and to assess the consistency of this effect in PC12 cells.Methods Mice were divided into FAC treatment group and control group.OB tissue was collected,and RNA sequencing(RNA-seq)was per-formed to identify differentially expressed genes associated with the synaptic vesicle cycle pathway;qPCR was used to validate the changes in the expression of the genes such as Slc17a8,Slc6a12,and Unc13c,which was verified in PC12 cells.Results RNA-seq showed that FAC treatment significantly downregulated the expression of the genes associated with the synaptic vesicle cycle pathway in OB tissue.The results of qPCR validation further confirmed significant downregulation of the Slc17a8,Slc6a12,and Unc13c genes in both OB tissue(t=3.184-4.285,P<0.05)and PC12 cells(t=4.540-13.350,P<0.05),which was consistent with the results of RNA-seq.Conclusion Iron treatment can downregulate the expression of the genes associated with the sy-naptic vesicle cycle pathway in OB tissue and PC12 cells,suggesting that iron may impair the normal function of the synaptic vesicle cycle pathway.关键词
