依布硒在真菌性角膜炎中的体外抗真菌及抗炎作用
In vitro anti-fungal and anti-inflammatory effects of ebselen in fungal keratitis

Abstract
Objective To investigate the in vitro anti-fungal and anti-inflammatory effects of ebselen(EB)in fungal kerati-tis.Methods The minimal inhibitory concentration(MIC)of EB on Aspergillus fumigatus was determined.Calcium fluores-cent white(CFW)staining and propidium iodide(PI)staining were used to observe the effect of EB at various concentrations(0,8,16,32 mg/L)on the mycelial growth and cell membrane of A.fumigatus;HE staining was used to observe the ability of A.fumigatus spores to adhere to human corneal epithelial cells(HCECs);CCK-8 assay was used to observe the effect of different concentrations of EB on the viability of mouse macrophages(RAW264.7).Quantitative real-time PCR was used to measure the changes in the mRNA expression levels of the inflammatory cytokines IL-1β,IL-6 and TNF-α in RAW264.7 macrophages stimu-lated by inactivated A.fumigatus hyphae and treated by EB.Results MIC results showed that EB inhibited the growth of A.fumigatus.After 24 hours of culture,EB started to suppress A.fumigatus at a concentration of 4 mg/L in a concentration-de-pendent manner,and when the concentration of EB reached 32 mg/L or higher,it showed a significant inhibitory effect on A.fu-migatus which lasted to 48 hours(F=74.0-217.8,P<0.001).CFW staining showed a gradual reduction in the density of A.fu-migatus hyphae with the increase in the concentration of EB,and PI staining showed that EB effectively disrupted fungal cell mem-branes.After EB treatment,there was a reduction in the number of spores adhering to the surface of HCECs(t=5.282,P<0.001).EB solutions with a concentration of ≤64 mg/L did not affect the viability of RAW264.7 macrophages(P>0.05).Further-more,EB inhibited A.fumigatus-induced inflammatory response in RAW264.7 macrophages and downregulated the mRNA ex-pression levels of IL-1β,IL-6,and TNF-α(F=100.9-168.3,P<0.001).Conclusion EB can inhibit the growth of A.fu-migatus,reduce the adhesion ability of its cell membrane and spores,and suppress inflammatory response in macrophages induced by A.fumigatus.关键词
