Abstract

"Introduction of South Bamboo to the North"is a strategically significant forestry project in China.The aquaporin(AQP)family members of Phyllostachys glauca were identified at whole-genome level,and their expression patterns were analyzed to reveal their functions in the process of responding to salt stress,providing a theoretical basis for the selection of bamboo species for"Introduction of South Bamboo to the North"in alkali area.A total of 62 AQP genes were identified in the genome of Ph.glauca.Collinearity analysis revealed 36 collinear gene pairs within PgAQPs and 72 collinear gene pairs with AQP genes in Oryza sativa,involving 44 PgAQPs and 26 OsAQPs.Phylogenetic analysis indicated PgAQPs were classified into four subfamilies:TIP,PIP,NIP and SIP.According to subcellular localization prediction,PgAQPs were mainly localized to the plasma membrane and vacuole membrane.Conserved motif analysis showed 20 motifs were identified in PgAQPs,with motif 3 conserved in all the members.Promoter analysis of PgAQPs revealed that multiple responsive elements related to hormone signaling and abiotic stress were found.RNA-seq data analysis demonstrated that nine PgAQPs were expressed across all the examined tissues,14 were not detected or were expressed at negligible levels in all tissues,and the remaining members showed tissue-specific expression patterns.A total of 15 PgAQPs were selected for qRT-PCR analysis under salt stress,of which five genes displayed an expression pattern of initial up-regulation followed by down-regulation,six genes exhibited initial down-regulation followed by up-regulation,and the remaining genes were persistently down-regulated throughout the treatment.These results suggest that they may have different potential roles in the salt-stress response of Ph.glauca,which be potential gene resources for resistance breeding of bamboo.
