A Previous Undescribed Dihydroxybenzoic Acid-triterpenoid Hybrids from Ganoderma lucidum

Abstract
Objective Isolation and characterization of chemical components in Ganoderma lucidum and evaluation of their biological activity against triple-negative breast cancer.Methods 500.0 kg of dried seeds of Ganoderma lucidum were taken,crushed and extracted with 80%ethanol under reflux,and concentrated under reduced pressure to obtain the crude extract was extracted with equal volume of ethyl acetate for 4 times,and concentrated under reduced pressure to obtain the ethyl acetate part of the extract was 11.5 kg.The compounds were then analyzed by a variety of techniques,such as MCI gel CHP 20P,RP-18,Sephadex LH-20,silica gel column chromatography,preparative thin-layer chromatography,semi-preparative thin-layer chromatography,and other column chromatography,and semi-preparative thin layer chromatography,and combined with the guidance of LC-MS/DAD and other techniques to selectively isolate compounds with novel struc-tures.The structural identification and stereoconformational determination of the compounds were also carried out by high resolution mass spectrometry(HRMS),nuclear magnetic resonance spectroscopy(NMR)(1D and 2D NMR),and chemical methods.Results Five compounds including a previously undescribed dihydroxybenzoic acid-triterpenoid hybrids were isola-ted from the 80%ethanol extract of Ganoderma lucidum by MCI Gel CHP-20P,RP-18,and sephadex LH-20 column chro-matography.They were identified as ganolucinin T(1),11α-hydroxy-3,7-dioxo-5α-lanosta-8,24(E)-dien-26-oic acid(2),3,7,11-trioxo5a-lanosta-8,24(E)-dien-26-oic acid(3),ganodermic acid T-Q(4),and ganorbimormin G(5)by spectro-scopic and chemical methods as well as by comparision with literature data.Finally,the preliminary evaluation of CCK-8 detection showed that compound 1 had anti-triple-negative breast cancer activity.Conclusion Five compounds were isolated from Ganoderma lucidum,among which compound 1 was new and showed anti-triple-negative breast cancer activity.关键词
