硅胶塞和牛皮纸两种三角瓶封口方法对微生物培养基的影响
Influence on Microbial Culture Medium Sealing with Silica Gel Plug and Kraft Paper

Abstract
Microbial culture medium must be sterilized before use to avoid microbial contamination,and then the sterility test was performed on medium uninoculated microorganisms overnight and showed no detectable microbial growth to check the ste-rility effect.The sealing method of the culture medium is crucial for the maintaining of sterility.This experiment compared the sterility effect maintaining between traditional sealing method with kraft paper and the currently commonly used sealing method with silica gel plug of two types of microbial culture media,potato-dextrose agar medium(PDA)and beef extract peptone solid culture medium(nutrient agar medium).The results showed that:(1)All the media uninoculated microorganisms sealed with kraft paper and silica gel plug were clear after 2 d of culture in an incubator at 28℃;(2)All the media sealed with silica gel plug were overgrown with microorganisms or white velum after 10 d of culture,while the media sealed with kraft paper remained clear and sterile;(3)The media sealed with kraft paper remained clear and sterile after 110 d of culture,and then the media were overgrown with microorganisms after 14 d of culture follow-ing the replacement of kraft paper with silicone stopper.These results indicated that the sterility effect maintaining of culture medium by traditional sealing method with kraft pa-per is better than the currently commonly used sealing meth-od with silica gel plug,and the medium sealed with kraft pa-per can still be used for microbial cultivation after long-term preservation.关键词
