摘要

Abstract

Objective It is to observe the effects of Jin Kui Shen Qi Pill on kidney tissue,mitochondrial respiratory chain complex activities,and content of adenosine triphosphate(ATP)in aging SD rats,and to explore its mechanism in improving energy metabolism disorders in aging rats to delay aging.Methods Sixty 2-month-old SD rats were taken,in which 12 rats were randomly selected as young group,and the remaining rats were naturally fed until 15 months of age,then they were randomly divided into aged group,low-dose,medium-dose,and high-dose groups of Jin Kui Shen Qi Pill,with 12 rats in each group.The low-dose,medium-dose,and high-dose groups of Jin Kui Shen Qi Pill were intragastrically administered Jin Kui Shen Qi Pill at doses of 0.54 g/(kg·d),1.08 g/(kg·d),and 2.16 g/(kg·d),respectively,all once daily,continuously treated for 3 months.Then the rats were sacrificed and their kidney tissues were collected for β-galactosidase staining and HE staining to observe histological changes;the mitochondria were isolated from kidney tissues to detect activities of mitochondrial respiratory chain complex Ⅰ,Ⅱ,Ⅲ,and Ⅳ by spectrophotometry,and to measure the content of mitochondrial ATP by chemiluminescence.Results Compared with the young group,the kidney tissue of rats in the aged group was significantly aging,with widespread damage in renal cortex,extensive cellular edema,ballooning de-generation,and mild interstitial inflammatory infiltration;the activities of mitochondrial respiratory chain complexⅠ,Ⅱ,Ⅲ,Ⅳ and the content of mitochondrial ATP in renal tissue were all reduced(all P<0.05).Compared with the aged group,the kidney tissue aging and damage were significantly alleviated in all Jin Kui Shen Qi Pill groups;the activities of mitochondrial respiratory chain complex Ⅰ,Ⅲ,and the content of mitochondrial ATP in renal tissue in the medium-dose and high-dose groups of Jin Kui Shen Qi Pill,and the activities of mitochondrial respiratory chain complexⅡ,Ⅳin all Jin Kui Shen Qi Pill groups were significantly increased(all P<0.05);the activities of mitochondrial respiratory chain com-plex Ⅰ,Ⅱ,Ⅲ,Ⅳ,and the content of mitochondrial ATP in renal tissue in the high-dose group of Jin Kui Shen Qi Pill were significantly higher than those in the medium-dose and low-dose groups(all P<0.05).Conclusion Jin Kui Shen Qi Pill can alleviate kidney tissue damage in aging rats,the effects may be achieved potentially by regulating mitochondrial re-spiratory chain complex activities to improve mitochondrial respiratory function and energy metabolism.
