Effect of Yijing decoction on ovarian angiogenesis in rats with reduced ovarian reserve function via regulating SIRT3/HIF-1α/PDK1 signaling pathway

Abstract
Objective It is to investigate the effect of Yijing decoction on ovarian angiogenesis and oocyte secretory fac-tors in rats with diminished ovarian reserve(DOR)via regulating silent mating-type information regulation 3(SIRT3)/hy-poxia inducible factor-1α(HIF-1α)/pyruvate dehydrogenase kinase 1(PDK1)signaling pathway.Methods Fifty-five SD female rats were taken,in which 10 rats were randomly selected as normal group,while the remaining rats were adminis-tered a tripterygium glycosides tablets suspension by gavage to establish DOR models.The 40 successfully modeled rats were randomly divided into 4 groups:model group,low-dose Yijing decoction group,high-dose Yijing decoction group,and high-dose Yijing decoction+3-TYP group,with 10 rats in each group.The low-dose and high-dose groups of Yijing decoc-tion were treated with Yijing decoction containing 5.895 g and 11.79 g crude drug by gavage,respectively,the high-dose Yi-jing decoction+3-TYP group was treated with Yijing Decoction containing 11.79 g crude drug combined with 3-TYP 50 mg/kg,the normal and model groups were given an equal volume of physiological saline,all once daily,continuously administered for 14 days.The serum levels of estradiol(E2),anti Mullerian hormone(AMH),luteinizing hormone(LH),and follicle stimulating hormone(FSH)were measured by ELISA,the contents of reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)in ovarian tissue were measured by reagent kits,the pathological morphology of ovarian tissue was observed by HE staining,ovarian angiogenesis was detected by im-munofluorescence staining,while the protein expressions of growth differentiation factor-9(GDF-9),bone morphogenetic protein-15(BMP-15),vascular endothelial growth factor(VEGF),SIRT3,HIF-1α,and PDK1 in ovarian tissue were de-tected by Western blot analysis.Results Compared with the normal group,the estrous cycle of rats in the model group was significantly prolonged(P<0.05),and ovarian tissue was damaged;the serum levels of E2 and AMH,contents of SOD and GSH-Px,CD31 fluorescence expression values,and relative protein expressions of GDF-9,BMP-15,VEGF,SIRT3,HIF-1α,and PDK1 in ovarian tissue were significantly reduced(all P<0.05),while the serum levels of FSH and LH,contents of ROS and MDA in ovarian tissue,and ovarian granulosa cell apoptosis rate were significantly increased(all P<0.05).Compared with the model group,the estrous cycle of rats in the low-dose Yijing decoction group,high-dose Yijing decoction group was significantly prolonged(P<0.05),and ovarian tissue damage was significantly alleviated;the serum levels of E2 and AMH,contents of SOD and GSH-Px,CD31 fluorescence expression values,and relative protein expres-sions of GDF-9,BMP-15,VEGF,SIRT3,HIF-1α,and PDK1 in ovarian tissue were significantly increased(all P<0.05),while the serum levels of FSH and LH,contents of ROS and MDA in ovarian tissue,and ovarian granulosa cell ap-optosis rate were significantly decreased(all P<0.05).3-TYP inhibited Yijing decoction in improving ovarian reserve function of DOR rats,the differences in all the above indicators were statistically significant when compared with the high-dose Yijing decoction group(all P<0.05).Conclusion Yijing decoction can alleviate ovarian tissue damage and improve ovarian reserve function in DOR rats maybe through activating SIRT3/HIF-1α/PDK1 signaling pathway,thereby inhibiting oxidative stress and ovarian granulosa cell apoptosis,and promoting ovarian angiogenesis and expression of oocyte secretion factors.关键词
