摘要

Abstract

Objective It is to discuss the mechanism of decoction for resolving turbidity and removing toxin in impro-ving renal function in 5/6 nephrectomy rats base on the silent mating-type information regulation 2(SIRT1)/FoxO1 signa-ling pathway.Methods Fifty-six rats were randomly selected from 67 SD rats and subjected to a 5/6 nephrectomy to estab-lish models of chronic renal failure(CRF),while the remaining 11 rats were as the sham operation group.The successfully modeled rats were randomly divided into renal failure group and 4 medicated groups,with 9 rats in each group.The losartan potassium group was treated with losartan potassium 4.5×10-3g/kg by gavage,the low,medium,and high-dose groups of decoction for resolving turbidity and removing toxin were treated with decoction for resolving turbidity and removing toxin 1.175 g/kg,2.35 g/kg,and 4.7 g/kg by gavage,respectively,the sham operation group and renal failure group were giv-en normal saline by gavage.After 8 weeks of continuous gavage,blood was collected from the abdominal aorta to measure renal function indicators[serum creatinine(SCr),blood urea nitrogen(BUN)]by biochemical methods,and oxidative stress indicators[superoxide dismutase(SOD),malondialdehyde(MDA),reactive oxygen species(ROS)]by immu-nosorbent assay(ELISA).The animals were sacrificed and their renal tissue was taken to observe renal histopathology and collagen fiber deposition by HE staining and Masson's trichrome staining,and to detect positive expression of collagen I in renal tissue by immunofluorescence,protein expressions of SIRT1 and FoxO1 in renal tissue by Western blot analysis.Re-sults Compared with the sham operation group,the serum levels of SCr,BUN,MDA,and ROS were significantly increased(all P<0.05),while serum level of SOD was significantly reduced in the renal failure group(P<0.05);partial glomeru-lar atrophy was observed,along with loss of the brush border and vacuolar degeneration in the renal tubules,as well as ac-cumulation of collagen fibers in the periglomerular and interstitial areas;the fluorescence intensity of Collagen I was in-creased in renal tissue,while the relative protein expressions of SIRT1 and FoxO1 were significantly reduced(all P<0.05).Compared with the renal failure group,the serum levels of SCr,BUN,MDA,and ROS were significantly de-creased(all P<0.05),while serum levels of SOD were significantly induced in the medicated groups(all P<0.05);the pathological damage in renal tissue was obviously alleviated;the fluorescence intensities of Collagen I in renal tissue were decreased,while the relative protein expressions of SIRT1 and FoxO1(except for FoxO1 in the low-dose group of decoction for resolving turbidity and removing toxin)were significantly increased(all P<0.05).Conclusion Decoction for resol-ving turbidity and removing toxin can effectively improve renal function and inhibit renal fibrosis in rats after 5/6 nephrecto-my,the mechanism which may be related to activate SIRT1/FoxO1 signal pathway,thereby to inhibit oxidative stress injury.
