Potential mechanism of decoction for replenishing Qi,resolving stasis and removing toxin in the treatment of rats with acute respiratory distress syndrome based on transcriptomics

摘要

Abstract

Objective It is to explore the mechanism of decoction for replenishing Qi,resolving stasis and removing toxin in the treatment of rats with acute respiratory distress syndrome(ARDS)based on transcriptomics.Methods Fifteen male SD rats were randomly divided into 3 groups.the group of decoction for replenishing Qi,resolving stasis and removing toxin was given decoction for replenishing Qi,resolving stasis and removing toxin 12.2 g/kg by gavage,the blank group and model group were given an equal volume of distilled water by gavage,all once daily,continuously administered for 7 days.After the final gavage on the 7th day,the group of decoction for replenishing Qi,resolving stasis and removing toxin and model group were given lipopolysaccharide(LPS)2 mg/kg once by tail vein injection to establish models of ARDS.The rats were sacrificed at 16 hours after modeling,their lung tissue macroscopic appearance was observed,histopathology was observed by HE staining,and the contents of interleukin-10(IL-10)and interleukin-6(IL-6)in lung tissue were measured by ELISA,the potential mechanisms of decoction for replenishing Qi,resolving stasis and removing toxin in the treatment of ARDS were analyzed by Transcriptomic and bioinformatics analysis.Results Significant pulmonary tissue dam-age was found in rats of the model group;the content of IL-10 in lung tissue was significantly lower,while the content of IL-6 in lung tissue was significantly higher than those in the blank group(both P<0.05).The pulmonary tissue damage in the group of decoction for replenishing Qi,resolving stasis and removing toxin was milder than that in the model group;the content of IL-10 in lung tissue was significantly higher,while the content of IL-6 in lung tissue was significantly lower than those in the blank group(both P<0.05).Transcriptomic analysis revealed 45 differentially expressed genes across the three groups,with secretory phosphoprotein 1(Spp1)identified as the core gene.Kyoto Encyclopedia of Genes and Ge-nomes(KEGG)pathway enrichment analysis revealed that the primarily enriched pathways of therapy for replenishing Qi,resolving stasis and removing toxin were cytokine-cytokine interaction pathway,PI3K-Akt signaling pathway,Toll-like re-ceptor signaling pathway,focal adhesion signaling pathway,and extracellular matrix-cell surface receptor interaction path-way.Conclusion Therapy for replenishing Qi,resolving stasis and removing toxin can regulate inflammatory homeostasis and alleviate lung tissue damage in ARDS rats,and its mechanism may be related to the regulation of Spp1 and other gene expressions,thus to regulate signal pathways such as PI3K-Akt,TOLL-like receptors,focal adhesions,and extracellular matrix-cell surface receptor interaction pathway.
