齐墩果酸调节IL-6/JAK2/STAT3信号通路对银屑病大鼠皮肤损伤的影响
Effect of oleanolic acid on skin injury in psoriasis rats by regulating IL-6/JAK2/STAT3 signaling pathway

Abstract
Objective:To investigate effect of oleanolic acid(OA)on skin injury in psoriasis(PsO)rats by regulating IL-6/Janus kinase 2(JAK2)/signal transducer and activator of transcription-3(STAT3)signaling pathway.Methods:Rats were separated into control group,PsO group,low,medium and high-dose OA groups(OA-L,OA-M,OA-H)and rIL-6 group(IL-6/JAK2/STAT3 signaling pathway activator).Except for control group,all other rats were induced into PsO models by imiquimod cream.After drug intervention,local skin injury was observed,and severity index PASI score of rat PsO area was evaluated.HE staining was applied to observe pathological changes in skin.ELISA was applied to detect levels of TNF-α and IL-6 in serum.qRT-PCR was applied to detect mRNA expressions of IL-6,JAK2 and STAT3.Western blot was applied to detect expressions of IL-6/JAK2/STAT3 pathway proteins.Results:Compared with control group,PASI score,epidermal thickness,inflammatory cell infiltration score,TNF-α,IL-6 levels,IL-6,JAK2,STAT3 mRNA,and IL-6,p-JAK2/JAK2,p-STAT3/STAT3 protein expressions in PsO group increased(P<0.05).Com-pared to PsO group,PASI score,epidermal thickness,inflammatory cell infiltration score,TNF-α,IL-6 levels,IL-6,JAK2,STAT3 mRNA,and IL-6,p-JAK2/JAK2,p-STAT3/STAT3 protein expressions were reduced in OA-L,OA-M and OA-H groups,and showed concentration dependence(P<0.05).IL-6/JAK2/STAT3 signaling pathway activator rIL-6 obviously reduced improvement effect of OA on skin injury in PsO rats(P<0.05).Conclusion:OA has effects of inhibiting inflammatory response and improving skin injury in PsO rats,and its mechanism is related to inhibiting activation of IL-6/JAK2/STAT3 signaling pathway.关键词
