Platycodin D inhibits LPS induced oxidative stress and neuroinflammation in astrocytes through Nrf2 and NF-κB

Abstract
Objective:To explore the effects of platycodin D on lipopolysaccharide(LPS)induced astrocyte oxidative stress and cellular inflammatory response,as well as roles of Nrf2 and NF-κB pathways.Methods:Isolation and identification of astrocytes from the certex of 2-day old Wistar rats.With 1 μmol/L LPS induced astrocytes to establish a cellular inflammatory model,which was divided into control group,LPS group,and LPS+platycodon D(1,10,100 μmol/L)groups.Cell survival rate was measured by MTT assay,NO content was measured using Griess reaction,lipid peroxide malondialdehyde(MDA)and superoxide dismutase(SOD)contents were detected by reagent kits.Western blot was used to detect expression levels of cyclooxygenase-2(COX-2),inducible nitric oxide synthase(iNOS),IL-1β,IL-17,chemokine ligand 2(CCL2),chemokine C-X-C motif ligand 13(CXCL13),Nrf2,kelch like ECH associated protein 1(Keap1),NF-κB1,NF-κB2,IKBκB and V-rel reticuloendothelial virus oncogene homolog A(RELA).Results:LPS induced astrocytes showed a decrease in survival rate,an increase in NO production,an increase in MDA content,a decrease in SOD activity,up-regulation of COX-2,iNOS,IL-1β,IL-17,CCL2,CXCL13,Keap1,NF-κB1,NF-κB2,IKBκB and RELA expressions,while down-regulation of Nrf2 expression(P<0.05).Survival rate of astrocytes induced by LPS pretreatment with different concentrations of platycodon D(1,10 and 100 μmol/L)was increased,with less NO production,decreased MDA content,increased SOD activity,down-regulated expressions of COX-2,iNOS,IL-1β,IL-17,CCL2,CXCL13,Keap1,NF-κB1,NF-κB2,IKBκB and RELA,while up-regulated expression of Nrf2(P<0.05).Conclusion:Platycodin D has inhibitory effects on LPS induced oxidative stress and neuroinflammation in astrocytes,and exerts its effects through Nrf2 and NF-κB signaling.Platycodin D is a potential drug for development of neuroimmune related disease treatments and prevention.关键词
