肉桂醛调节JAK2-STAT3信号通路对牙周炎大鼠牙周组织损伤的影响
Effect of cinnamaldehyde on periodontal tissue damage in periodontitis rats by regulating JAK2-STAT3 signaling pathway

Abstract
Objective:To investigate effect of cinnamaldehyde(CA)on periodontal tissue damage in periodontitis rats by regulating JAK2-STAT3 signaling pathway.Methods:Rats were grouped into Control group,Model group,CA low-dose(CA-L)group,CA high-dose(CA-H)group and CA-H+Colivelin(JAK2 activator)group randomly.A periodontitis model was established by ligating neck of maxillary first molar of rats with silk thread.ELISA was applied to detect levels of TNF-α,IL-10 and IL-1β in rat serum.Micro-CT testing was used to analyze loss of alveolar bone.Tartrate resistant acid phosphatase(TRAP)staining was used to observe osteoclastogenesis.HE staining was applied to observe morphology of periodontal tissue.Western blot was used to detect levels of OPG,RANKL and JAK2-STAT3 signaling pathway proteins in periodontal tissue.Results:Compared with Control group,rats in Model group showed severe bone loss and obvious damage to periodontal tissue,CEJ-ABC distance,serum TNF-α and IL-1β levels,periodontal histopathological score,osteoclast counts,RANKL,p-JAK2/JAK2 and p-STAT3/STAT3 protein expressions elevated,bone volume fraction,serum IL-10 level,and OPG protein expression in periodontal tissue were decreased(P<0.05).Compared with Model group,bone loss and periodontal tissue damage were obviously reduced in CA-L and CA-H groups,CEJ-ABC distance,serum TNF-α and IL-1β levels,periodontal histopathological score,osteoclast count,RANKL,p-JAK2/JAK2 and p-STAT3/STAT3 protein expressions were decreased,bone volume fraction,serum IL-10 level,and OPG protein expression in periodontal tissue were increased(P<0.05).Colivelin was able to reduce improvement of periodontal tissue damage caused by CA in periodontitis rats(P<0.05).Conclusion:CA can reduce bone resorption,inflammatory response and osteoclastogenesis in periodontitis rats,alleviate periodontal tissue damage,and may be related to inhibition of JAK2/STAT3 signaling pathway.关键词
