摘要

Abstract

AIM:To overcome the limitations of sodium alginate hydrogel(SAH),including its slow degradation and limited bioactivity in wound heal-ing applications,we developed a functionalized hy-drogel(SAQM)incorporating Mg²⁺ and quercetin(Que).We further conducted a systematic evalua-tion of its roles and underlying molecular mecha-nisms in modulating neutrophil-mediated inflam-matory responses and oxidative stress.METHODS:SAH and SAQM were prepared via ionic crosslink-ing induced by CaCl2.Morphology was character-ized by SEM,mechanical properties were analyzed via rheometry,and component release was quanti-fied using ICP-AES/UV spectrophotometry.In vitro evaluation employed a dHL60 neutrophil model stimulated with LPS.Inflammatory markers and NF-kB pathway activation were assessed by RT-qPCR and Western blot,while ROS generation was mea-sured via flow cytometry.RESULTS:In vitroexperi-ments demonstrated sustained release of Mg2+and Que from SAQM.SEM and rheological analyses re-vealed comparable gelation and microstructure be-tween SAQM and SAH,but SAQM showed signifi-cantly reduced swelling and enhanced degradation rates.Biocompatibility studies confirmed negligible cytotoxicity toward neutrophils.In LPS-induced in-flammation,SAQM markedly suppressed pro-in-flammatory cytokine expression and ROS produc-tion.Compositional analysis indicated Mg2+primari-ly mediated anti-inflammatory effects,while Que contributed to antioxidant activity.Notably,SAQM treatment inhibited LPS-induced activation of the canonical NF-kB pathway,suggesting its anti-inflam-matory mechanism involves NF-kB suppression.CONCLUSION:SAQM combines improved degrad-ability with dual anti-inflammatory and antioxidant properties,representing a promising immunomodu-latory hydrogel dressing for clinical applications.
