溃结宁膏穴位敷贴调控内质网应激介导的IRE1α/JNK通路对溃疡性结肠炎大鼠结肠上皮细胞凋亡的影响
Exploring the Impact of Kuijiening Ointment Acupoint Application on Colonic Epithelial Cells in Ulcerative Colitis Rats Based on Endoplasmic Reticulum Stress-Mediated IRE1α/JNK Apoptotic Pathway

Abstract
Objective:To observe the therapeutic effect of Kuijiening Ointment acupoint application on ulcer-ative colitis(UC)rats under a state of"deficiency and stasis"and its impact on the endoplasmic reticulum stress(ERS)-mediated IRE1α/JNK pathway,thereby exploring the mechanism of action of this treatment for UC.Meth-ods:SD rats were randomly divided into a blank group(n=10)and a modeling group(n=50).The UC model un-der a state of"deficienoy and stasis"was induced using adenine and senna leaf gavage followed by 2,4,6-trinitro-benzene sulfonic acid enema.After modeling,the rats were further divided based on body weight into model group,non-acupoint application group,acupoint without application group,acupoint application group,and salicylazosulfa-pyridine(SASP)group,each receiving corresponding treatments for 2 weeks.Disease Activity Index(DAI)and Co-lon Mucosal Damage Index(CMDI)scores were calculated.Histopathological changes in colon tissues were ob-served after HE staining.ELISA was used to measure FasL and IL-8 levels in colon tissue homogenates.TUNEL assay was performed to determine the apoptosis rate of colonic epithelial cells.Western blotting was used to detect protein expression levels of GRP94,p-IRE1α,p-JNK,Bax,Bcl-2,and Cleaved Caspase-3 in colon tissues.Re-sults:Compared with the blank group,the model group showed reduced body weight,irregular colonic mucosa mor-phology,significant ulcers,and inflammatory cell infiltration.DAI and CMDI scores increased(P<0.01),as were FasL and IL-8 levels in colon tissue homogenates(P<0.01).Apoptosis rates of colonic epithelial cells increased(P<0.01),along with elevated protein expression levels of GRP94,p-IRE 1α,p-JNK,Bax,and Cleaved-Caspase-3(P<0.01)and decreased BCL2 protein expression(P<0.01).Compared with the model,non-acupoint application,and acupoint without application groups,both the acupoint application group and the SASP group exhibited in-creased body weight,reduced pathological damage to the colonic mucosa,lower DAI and CMDI scores(P<0.01),decreased FasL and IL-8 levels(P<0.01),reduced apoptosis rates(P<0.01),and downregulated protein expres-sion levels of GRP94,p-IRE 1α,p-JNK,Bax,and Cleaved Caspase-3(P<0.01),with upregulated Bcl-2 protein expression(P<0.01).There were no statistically significant differences between the acupoint application group and the SASP group in terms of various scores and apoptosis rates(P>0.05).Conclusion:Kuijiening Ointment acupoint application may treat UC by regulating the ERS-mediated IRE1α/JNK pathway,reducing excessive apop-tosis of intestinal epithelial cells.关键词
