摘要

Abstract

Objective:To study how Huanglian Jiedu Decoction affects the damage of periodontal tissues in rats with periodontitis through autophagic pathway and its mechanism of action.Methods:50 SD rats were randomly divided into 5 groups:blank control group,model group,low-dose and high-dose Huanglian Jiedu Decoction groups,and high-dose+autophagy inhibitor 3-methyladenine(combination)group.Except for the blank control group,periodontitis models were established in the other groups.The inflammatory state of periodontal tissues was evaluated.Micro-CT tomography was used to observe alveolar bone resorption.Enzyme-linked immunosorbent as-say(ELISA)was used to detect serum interleukin-1β(IL-1 β),tumor necrosis factor-α(TNF-α),and interferon-γ (IFN-γ).HE staining was used to observe pathological changes of periodontal tissues.Immunohistochemical stain-ing was used to observe the expression of IL-1β in periodontal tissues.Western blotting was used to detect the ex-pression changes of microtubule-associated protein 1 light chain 3(LC3),Beclin-1,and P62 proteins in periodon-tal tissues.Results:Compared with the control group,the model group showed gingival papilla defect,epithelial damage with ulceration,more inflammatory cells in the gingiva,disordered periodontal structure,partial fibrosis and alveolar bone resorption.The gingival index,gingival bleeding index,CEJ-ABC value,serum levels of IL-1 β,TNF-α,IFN-γ,and the expression levels of IL-1β and P62 proteins in periodontal tissues were increased(P<0.05),while the expression levels of LC3-Ⅱ/LC3-Ⅰ and Beclin-1 proteins in periodontal tissues were decreased(P<0.05).Compared with the model group,the low-dose and high-dose Huanglian Jiedu Decoction groups showed reduced alveolar bone damage and alleviated inflammation.The gingival index,gingival bleeding index,CEJ-ABC value,serum levels of IL-1β,TNF-α,IFN-γ,and the expression levels of IL-1 β and P62 proteins in periodontal tissues were decreased(P<0.05),while the expression levels of LC3-Ⅱ/LC3-Ⅰ and Beclin-1 pro-teins in periodontal tissues were increased(P<0.05).The autophagy inhibitor 3-MA could reverse the protective effect of Huanglian Jiedu Decoction on periodontal tissue damage in rats with periodontitis(P<0.05).Conclu-sion:Huanglian Jiedu Decoction may alleviate periodontitis in rats by activating the autophagic pathway and there-by inhibiting IL-1 β.
