Effect of Stellate Ganglion Catgut Embedding on Substance P and Th1/Th2 Cytokines in Asthmatic Rats

Abstract
Objective:To observe the effect of stellate ganglion catgut embedding on substance P(SP)and Th1/Th2 cytokines in asthmatic rats,and to explore its mechanism in treating asthma.Methods:40 SPF-grade male SD rats were randomly divided into 5 groups:blank group,model group,catgut embedding group,block group,and hormone group.The asthmatic rat model was established by ovalbumin sensitization and challenge.The catgut em-bedding group received stellate ganglion catgut embedding once;the block group received stellate ganglion block;the hormone group received budesonide atomization inhalation once a day for 7 consecutive days.The general con-dition and asthma behavioral score of rats were observed;HE staining was used to observe the lung tissue patholo-gy;Western blotting was used to detect the expression of SP protein in the stellate ganglion and lung tissue of rats;enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of SP,interferon-γ(IFN-γ),and inter-leukin-4(IL-4)in the serum and bronchoalveolar lavage fluid of rats.Results:Compared with the blank group,the model group showed symptoms such as wheezing,shortness of breath,and irritability with increased behavioral scores(P<0.01),obvious pathological damage of lung tissue,increased expression of SP protein in the stellate gan-glion and lung tissue(P<0.01),significantly increased levels of SP and IL-4 in serum and bronchoalveolar lavage fluid,and significantly decreased levels of IFN-γ and IFN-γ/IL-4 ratio in serum and bronchoalveolar lavage fluid.Compared with the model group,the catgut embedding group,block group,and hormone group showed improved symptoms such as wheezing,shortness of breath,and irritability with decreased behavioral scores(P<0.01),signifi-cantly improved pathological damage of lung tissue,decreased expression of SP protein in the stellate ganglion and lung tissue(P<0.01),significantly decreased levels of SP and IL-4 in serum and bronchoalveolar lavage fluid,and significantly increased levels of IFN-γ and IFN-γ/IL-4 ratio in serum and bronchoalveolar lavage fluid(P<0.01).Conclusion:Stellate ganglion catgut embedding can down-regulate the expression of SP protein,inhibit the levels of Th2-related cytokines,increase the levels of Th 1-related cytokines,and balance Th1/Th2,thereby reducing asth-matic lung inflammation and treating asthma.关键词
