Response of lotus seed germination to salt-alkali stress combination

Abstract
[Objective]The study aimed to investigate the changes in physiological and biochemical indicators of lotus seed germination under different concentrations of saline-alkali mixed stress,thereby providing a reference for enhancing the breeding of salt-alkali tolerant lotus seeds and promoting the rational development and utilization of saline-alkali land.[Method]Six widely used and nutritionally valuable lotus varieties('Jinfurong 1''Jinfurong 2''Taikong 1''Taikong 2''Chuzhoubailian'and'Baixianglian')were selected as experimental materials.Different saline-alkali mixed solutions(50,100,150 mmol·L-1,with a molar ratio of NaCl to NaHCO3 of 1∶1)were used to treat the seeds for germination,and the germination potential,germination rate,malondialdehyde(MDA)content,catalase(CAT)activity,peroxidase(POD)activity,superoxide dismutase(SOD)activity,and proline(Pro)content of the seeds under various saline-alkali concentrations were measured and analyzed.The fuzzy mathematical membership function method was used to comprehensively evaluate the salt-alkali tolerance of the six lotus varieties.[Result]Under different concentrations of saline-alkali treatment,the impact on the germination potential,germination rate,MDA content,CAT activity,POD activity,SOD activity,and Pro activity of the lotus seeds of each variety reached significant levels(P<0.05).The comprehensive analysis using the membership function method indicated that under low concentration(≤50 mmol·L-1)saline-alkali stress,the salt-alkali tolerance of the different varieties was in the order:'Taikong 2'>'Baixianglian'>'Chuzhoubailian'>'Taikong 1'>'Jinfurong 1'>'Jinfurong 2';under high concentration(100-150 mmol·L-1)saline-alkali stress,the salt-alkali tolerance was in the order:'Taikong 1'>'Chuzhoubailian'>'Taikong 2'>'Jinfurong 1'>'Jinfurong 2'>'Baixianglian'.[Conclusion]Under saline-alkali stress conditions,lotus seeds in the germination period could rapidly respond to saline-alkali stress by adjusting their own physiological state,but there were differences in the salt-alkali tolerance among different lotus varieties,mainly reflected in the dynamic changes of physiological and biochemical indicators.At the same time,the comprehensive evaluation using the fuzzy mathematical membership function method concluded that under high concentration saline-alkali stress,'Taikong 1'had the strongest salt-alkali tolerance.关键词
