Abstract

Otoliths are used as vital research materials in the disciplines of fish migratory ecology,conservation biology,and fishery resource conservation.Validating morphological differences between the left and right otoliths is critical for establishing specimen selection criteria and ensuring the comparability and objectivity of corresponding research outcomes.In this study,we aimed to elucidate whether morphological differences exist between bilateral otoliths in fish.Herein,for the first time,we used otoliths from both sides of Coilia nasus samples(including two ecotypes with long supermaxilla and short supermaxilla)to test potential morphological differences based on shape indices and geometric morphometric landmark features of sagittal otoliths.The results revealed no statistically significant differences in the shape indices,including length,height,area,and perimeter,indicating generally high similarity in outline structures.However,relative warp discriminant analysis revealed highly significant differences in geometric morphometric landmarks,primarily manifested in the main sulcus and secondary grooves,between bilateral otoliths.The intergroup discrimination success rate reached 100%,confirming distinct spatial configurations between the left and right otoliths.Based on these findings,we recommend that future otolith morphological studies must standardize protocols by consistently studying either the left or right otoliths to prevent potential asymmetry,thereby ensuring the comparability and objectivity of results.The validated differential characteristics of bilateral otoliths in C.nasus provide the urgently needed theoretical foundations for refining technical standards in otolith-related research in fishes.
