摘要

Abstract

To investigate the effects of salinity stress on the tissue structure,antioxidant capacity,and expression patterns of the Chloride Channel Protein 7 gene(ScCLC7)and Microsomal Glutathione S-Transferase 3 gene(ScMGST3)in Sinonovacula constricta,we exposed S.constricta to three different salinity levels(S20,S30,and S40)for 48 h.Subsequently,gill and hepatopancreas tissues were collected for preparing microscopic tissue sections,determining malondialdehyde(MDA)content and superoxide dismutase(SOD)activity,and analyzing the expression of ScCLC7 and ScMGST3 genes using quantitative reverse transcription polymerase chain reaction.Histological analysis revealed that both hypo-and hyper-saline conditions caused varying degrees of structural damage to gill and hepatopancreas tissues,with high salinity inducing relatively more severe damage.The MDA content and SOD activity in the gill tissue in the S40 group increased significantly,whereas there was little change in these parameters in the S20 group.In the hepatopancreas tissue,both hypo-and hyper-saline conditions increased the MDA content and SOD activity.Notably,MDA exhibited a common trend of initially increasing and subsequently decreasing under both hypo-and hyper-saline conditions.At the molecular level,ScCLC7 in gill was significantly up-regulated at 6 h of both hypo-and hyper-saline,and then recovered to the control level,while ScCLC7 in hepatopancreas showed a trend of decreasing and then increasing;both hypo-and hyper-saline up-regulated the expression of ScMGST3 in gill and hepatopancreas,but high salt affected gill to a greater extent than hepatopancreas,while the opposite effect is observed under low salt conditions.The results demonstrated that acute stresses with a 10‰ change in salinity significantly damaged the normal microstructure of the gills and hepatopancreas of S.constricta and also regulated their salinity acclimatization ability by altering the MDA content,SOD activity,and the expression of ScCLC7 and ScMGST3.This study provides a reference for analyzing the physiological responses and regulatory mechanisms of razor clams to acute salinity stress.
