Abstract

To explore the effect of exogenous trehalose solution on the low-temperature tolerance of flue-cured tobacco seedlings,the seedlings of the flue-cured tobacco variety Xiangyan No.7(at the 6-8 leaf stage)were used as the experimental materials.A controlled experiment design was adopted,with distilled water treatment and trehalose treatment set up.200 mL of distilled water and 15 mM of trehalose solution were applied externally for 24 hours,and then transferred to a light incubator for treatment at 25℃normal temperature and 4℃low temperature stress for 2 days.The phenotype was observed and analyze the changes in physiological and biochemical indices as well as the expression of stress-resistant genes.Phenotypic analysis showed that exogenous application of trehalose significantly alleviatedthe growth inhibition of tobacco caused by low temperature stress.Physiological and biochemical assay results showed that exogenous trehalose application significantly reduced malondialdehyde(MDA),relative electrical conductivity(EL),superoxide anion(OFR)and hydrogen peroxide(H2O2)contents of tobacco under low temperature stress;obviouslyincreased the activities of ascorbate peroxidase(APX),peroxidase(POD)glutathione(GSH)and catalase(CAT),increased contents of osmotic adjustment substance proline(PRO)and soluble protein(SP).Results of gene expression revealed that exogenous trehalose significantly upregulated the expression levels of these stress-responsive genes such as ERDs,CORs,RAB28,RD29A,and RD19 in tobacco seedlings under low-temperature stress.Comprehensive analysis shows that exogenous application of trehalose can enhance the tolerance of tobacco seedlings to low-temperature stress.
