黄精人参方对拟人不良生活方式致身心综合性疲劳小鼠模型的影响
Effect of Huangjing Renshen Formula on the Comprehensive Fatigue Model Mice Induced by Anthropomorphic Unhealthy Life-style
摘要
Abstract
[Objective]To explore the intervention effect of Huangjing Renshen Formula(HRF)on the comprehensive physical and mental fatigue model caused by anthropomorphic unhealthy lifestyle.[Methods]The mice were divided into normal control group,model control group,Compound Taurine Tablets(CTT)group(0.84 g·kg-1),and HRF low,medium and high dose groups(0.3,0.6,1.2 g·kg-1).Except for normal control group,all other groups were given corresponding drugs by gavage once a day for 4 weeks.At the same time,"sleep deprivation+cold water swimming+binding tail clamping method"was used to establish comprehensive physical and mental fatigue mouse model.During the experiment,the physical signs,exercise behavior and exhaustion swimming time of mice were observed,and the levels of energy substances such as muscle glycogen(MG),liver glycogen(LG),glucose(Glu);exercise metabolites lactate(LA),lactate dehydrogenase(LDH),urea nitrogen(BUN);oxidative stress indicators superoxide dismutase(SOD),malondialdehyde(MDA);hypothalamic neurotransmitters 5-hydroxytryptamine(5-HT),dopamine(DA)and norepinephrine(NE)were detected.Hematoxylin-eosin(HE)staining was used to observe the pathological changes in skeletal muscles of mice.[Results]Compared with normal control group,mice in model control group lost weight,shortened swimming time,significantly reduced grip strength,decreased levels of energy substances MG,LG and Glu,increased levels of metabolites LA,LDH and BUN,and had abnormal levels of 5-HT,DA and NE in the hypothalamus,indicating the successful establishment of comprehensive physical and mental fatigue mouse model.The HRF dosing groups significantly improved the weight loss of the mice(P<0.05),while the serum LA levels of HRF dosing groups significantly decreased,the serum GLU,MG levels,and SOD activity,as well as the hypothalamic DA level,significantly increased(P<0.01,P<0.05).The high and medium doses of HRF also enhanced the grip strength of the mice(P<0.01),extended the exhaustive swimming time(P<0.05),reduced the serum BUN level while increasing the LG level(P<0.05),and significantly prolonged the exercise distance and time in the open field test(P<0.05),with the HRF high dose group showing a significant increase in the number of grid crossings(P<0.05).[Conclusion]HRF can enhance the energy reserves of comprehensive physical and mental fatigue mice model,reduce metabolite accumulation,regulate neurotransmitter secretion disorders,and improve exercise endurance,thereby alleviating comprehensive physical and mental fatigue.关键词
