摘要

Abstract

[Objective]To investigate the differences in anti-hepatocellular carcinoma(anti-HCC)activity in vitro and secondary metabolites content of Tetrastigma hemsleyanum from different origins,in order to discover potential anti-HCC components in Tetrastigma hemsleyanum.[Methods]The contents of 12 compounds,total flavonoids and total polysaccharides in aqueous extracts of Tetrastigma hemsleyanum from eight origins including Fuzhou City in Fujian,Chongqing City,Hechi City in Guangxi,Enshi City in Hubei,Quzhou City in Zhejiang,Yongzhou City in Hu'nan,Shangrao City in Jiangxi and Dushan County in Guizhou were detected by high performance liquid chromatography(HPLC)with external standard single-point method or ultraviolet-visible(UV-Vis)spectrophotometry.The inhibition rates and migration rates of Tetrastigma hemsleyanum from different origins on HepG2 cells were compared by using cell counting kit-8(CCK-8)assay and Transwell migration assay separately.The spectrum-effect relationships between active components and inhibition/migration rates of HCC were analyzed by using correlation coefficients combined with orthogonal partial least squares discriminant analysis(OPLS-DA).[Results]The catechin content was highest in Fujian Tetrastigma hemsleyanum,while Zhejiang Tetrastigma hemsleyanum showed the highest content of characteristic component 2 kaempferol 3-O-β-D-glucoside-7-O-α-L-rhamnoside.Both Fujian and Zhejiang samples exhibited the highest total flavonoid and polysaccharide contents.The half maximal inhibitory concentration(IC50)of Zhejiang Tetrastigma hemsleyanum against HepG2 cells was 10.060 mg·mL-1.At the same crude drug concentration(5 mg·mL-1),inhibitory rate of Zhejiang Tetrastigma hemsleyanum(37.89%)against HepG2 cells was significantly higher than other producing areas,and migration rate of Zhejiang Tetrastigma hemsleyanum(45.6%)was significantly lower than other producing areas.Correlation analysis showed that gallic acid,catechin,total flavonoids and crude polysaccharides had a moderate or higher correlation with the inhibitory rate of HepG2 cells(r=0.8039,0.7179,0.7693,0.7576,P<0.05).[Conclusion]Including Fuzhou City in Fujian,Chongqing City,Hechi City in Guangxi,Enshi City in Hubei,Quzhou City in Zhejiang,Yongzhou City in Hu'nan,Shangrao City in Jiangxi and Dushan County in Guizhou,Tetrastigma hemsleyanum from Zhejiang showed the best in vitro anti-HCC activity with the same crude drug concentration,moreover,gallic acid,catechin,total flavonoids and crude polysaccharides could be the potential anti-HCC components of Tetrastigma hemsleyanum.
