通络兰白饮通过激活Nrf2/HO-1/NQO1信号通路减轻肾病综合征大鼠肾纤维化
Tongluo Lanbai Yin alleviates renal fibrosis in rats with nephrotic syndrome by activating the Nrf2/HO-1/NQO1 signaling pathway

Abstract
Objective To investigate whether Tongluo Lanbai Yin alleviates renal fibrosis in nephrotic syndrome(NS)rats by acti-vating the Nrf2/HO-1/NQO1 signaling pathway.Methods Sprague-Dawley rats were divided into five groups:normal control(control),nephrotic syndrome model(NS),low-dose Tongluo Lanbai Yin(L-TL),high-dose Tongluo Lanbai Yin(H-TL),and high-dose Tongluo Lanbai Yin+Nrf2 inhibitor ML385(H-TL+ML385).Urine protein,serum creatinine(Scr),blood urea nitrogen(BUN),albumin(Alb),cholesterol(CHOL),and triglycerides(TG)were measured in all groups.Superoxide dismutase(SOD)activity and the malondialdehyde(MDA)levels were assessed in renal tissue.Renal cell apoptosis was detected by TUNEL staining.Renal pathology was evaluated by HE staining.The expression levels of the Nrf2/HO-1/NQO1 signaling pathway and extracellular matrix deposition-related proteins were measured in renal tissues by Western blotting.Results Compared with the control group,the NS group showed increased levels of urine protein,Scr,BUN,TG,CHOL,MDA,collagenⅠ,fibronectin,and laminin,accompanied by severe renal tissue injury and increased apop-tosis;conversely,Alb,SOD,and the Nrf2,HO-1,and NQO1 protein levels decreased(P<0.05).Compared with the NS group,the L-TL and H-TL groups exhibited decreased levels of urine protein,Scr,BUN,TG,CHOL,MDA,collagenⅠ,fibronectin,and laminin,leading to alleviated renal tissue injury,and reduced apoptosis;conversely,Alb,SOD,and the Nrf2,HO-1,and NQO1 protein levels increased(P<0.05).Compared with the H-TL group,the H-TL+ML385 group showed increased levels of urine protein,Scr,BUN,TG,CHOL,MDA,col-lagenⅠ,fibronectin,and laminin,as well as aggravated renal tissue injury and increased apoptosis;conversely,Alb,SOD,and the Nrf2,HO-1,and NQO1 protein levels decreased(P<0.05).Conclusion Tongluo Lanbai Yin may activate the Nrf2/HO-1/NQO1 signaling pathway to enhance antioxidant capacity in NS rats,thereby reducing renal fibrosis and improving renal pathology and function.关键词
