杜仲叶总黄酮提取物对MC3T3-E1成骨细胞生长的作用研究
Effects of total flavonoids from Eucommia ulmoides leaves on the growth of MC3T3-E1 osteoblasts

Abstract
To investigate the effects of flavonoids and flavonoids(Kaempferol,Quercetin,Rutin,Astragalin,Baicalein)from Eucommia ulmoides leaves on the growth of MC3T3-E1 osteoblasts,and to extract total flavonoids from Eucommia ulmoides leaves by ultrasound-assisted ethanol method.MC3T3-E1 osteoblasts were treated with total flavonoids from Eucommia ulmoides leaves at 10~400 μg/mL for 24 h.The cell survival rate was detected by CCK-8 method,and the optimal concentration was screened.Total flavonoids from Eucommia ulmoides leaves were used to intervene cells for 24,48,72 and 96 hours,and cell growth curves were plotted.MC3T3-E1 osteoblasts were treated with Kaempferol,Quercetin,Rutin,Astragalin and Baicalein,and the cell survival rate was detected by CCK8 method.The proliferation of MC3T3-E1 osteoblasts was promoted by the concentration of total flavonoids of 10~200 μg/mL for 24 hours,and the survival rate of MC3T3-E1 osteoblasts was 112.87%,especially that of 100 μg/mL(P<0.05).The proliferation of MC3T3-E1 osteoblasts was inhibited by 400 μg/mL,and the cell survival rate was 47.86%.Quercetin,Rutin,Astragalin and Baicalein can improve the survival rate of osteoblasts,kaempferol can inhibit the proliferation of MC3T3-E1 osteoblasts.Conclusion This study shows that Eucommia ulmoides leaves are a good Chinese medicinal material for preventing and treating Osteoporosis,and the flavonoids extract from Eucommia ulmoides leaves can significantly improve the activity of MC3T3-E1 osteoblasts.Different flavonoids play different roles in bone formation and bone reconstruction,and have broad prospects for development and utilization.关键词
