Abstract
To investigate the effect of refined"Ziqisu"compound polysaccharide(C-APS)on the apoptosis of RAW264.7 macrophages,different concentrations of C-APS were added simultaneously with the apoptosis inducer 5-fluorouracil(5-FU)to induce cell apoptosis.Methods including the CCK-8 assay,Hoechst33342 staining,and Annexin V/PI double staining were employed,with macrophage morphology,proliferation,apoptosis,mitochondrial membrane potential,and apoptotic signaling pathways(PI3K-AKT,IRE1-JNK)used as evaluation indicators for the anti-apoptotic effect of C-APS.The results showed that C-APS at 78~5 000 μg/mL could maintain cell viability above 100%,while 5-FU(30 μmol/L)reduced the cell proliferation rate to 68.8%.Notably,the cell proliferation rate of the groups treated with C-APS(64~1 000 μg/mL)combined with 5-FU exceeded 100%,indicating that C-APS not only promotes macrophage growth but also resists 5-FU-induced macrophage apoptosis.Mitochondrial membrane potential detection revealed that C-APS could antagonize the 5-FU-induced decrease in mitochondrial membrane potential.Further studies demonstrated that C-APS enhances the activities of PI3K and AKT,while inhibiting JNK activity to resist macrophage apoptosis.In conclusion,"Ziqisu"compound polysaccharide exerts an anti-apoptotic effect on macrophages.关键词
