结直肠癌患者手术前后肠道菌群组成差异分析及功能预测
Composition differential analysis and functional prediction of gut microbiota before and af-ter surgery in colorectal cancer patients

Abstract
Objectives To investigate changes in the gut microbiota of colorectal cancer(CRC)patients before and after surgery using 16S rDNA sequencing technology,and to predict the function of differential microbiota and preliminarily ex-plore the impact of surgery on the microbiota,and provide intervention insights for reducing postoperative complications related to microbiota dysbiosis.Methods Fecal samples from patients undergoing radical CRC surgery were analyzed pre-operatively and postoperatively using 16S rDNA sequencing.A feature table was generated after data denoising and qual-ity control,and species abundance comparison was completed.Alpha and beta diversity analyses were employed to de-scribe species diversity between the two sample groups.Linear Discriminant Analysis Effect Size(LEfSe)was used to identify microbial taxa with significant intergroup differences.PICRUSt2 was utilized to predict the metagenomic func-tional composition of the microbial communities.Results At the genus level,the relative abundances of Akkermansia and Enterococcus were significantly increased,while the relative abundances of Prevotella_9 and Faecalibacterium were signifi-cantly decreased in the postoperative group.Alpha diversity analysis showed a significant decrease in the Shannon index postoperatively(P<0.05).Beta diversity analysis indicated significant separation between the preoperative and postopera-tive groups under Weighted/Unweighted Unifrac distance matrices(P<0.05).LEfSe analysis revealed Faecalibacterium and Enterococcus as significantly different between the two groups.Functional prediction suggested that genes related to amino acid metabolism,bacterial drug resistance,and transcription regulation pathways were primarily enriched in preop-erative samples,whereas genes related to carbohydrate metabolism were mainly enriched in postoperative samples.Con-clusion The diversity and richness of gut microbiota decrease in patients after CRC surgery.Postoperatively,the relative abundances of Akkermansia and Enterococcus increase significantly,while those of Prevotella_9 and Faecalibacterium de-crease significantly.Timely identification of microbiota disturbances and early regulation of the gut microecology may help prevent complications associated with postoperative gut microbiota dysbiosis.关键词
