Abstract

OBJECTIVE To evaluate sampling methods and clearance strategies for nasal bacteria in mice,providing reliable protocols for nasal microbiome research.METHODS Using nasal tissue(NT)bacterial culture counts as a reference,the suitability of pressing the nares(PN)and nasal lavage fluid(NLF)sampling methods for detecting nasal bacteria in mice was compared.Furthermore,NLF samples underwent bacterial culture and antibiotic susceptibility testing to assess the effectiveness of four clearance strategies:(1)free drinking of 5%glucose(GS)plus a four-antibiotic combination(4ABX),(2)free drinking of flavored water(FW)plus 4ABX,(3)free drinking of purified water,(4)intranasal instillation(i.n.)of a three-antibiotic combination(3ABX).RESULTS Compared with NLF and NT methods,the PN method yielded significantly higher numbers of Escherichia coli(P=0.041).No significant differences in bacterial profiles were observed between NT and NLF samples.Both the 3ABX i.n.group and the 5%GS+4ABX group achieved significantly higher nasal bacteria clearance rates than the FW+4ABX group(P<0.05).Primary residual colonies were antibiotic-insensitive.CONCLUSION NLF and NT sampling methods yield highly consistent bacterial profiles,making both suitable for mouse nasal microbiome studies.Local and systemic antibiotic treatments effectively reduce nasal bacterial loads;however,the persistence of resistant strains underscores the need for optimized clearance strategies.
