仿野生与GAP栽培穿心莲的品质特征及代谢谱比较
Comparative analysis of quality characteristics and metabolomic profiles of Andrographis paniculata between wild-simulated and GAP cultivation

Abstract
[Purpose]This study compared the agronomic traits,active components,and metabolites of Andrographis paniculata under wild-simulated(WS)cultivation and good agricultural practice(GAP)cultivation modes,aiming to provide a theoretical ba-sis and methodological support for the selection of cultivation methods and rational utilization of land resources.[Method]Agronom-ic trait investigation,component quantification,and non-targeted metabolomics were employed to compare the agronomic traits,ac-tive components,and metabolic profiles of dried whole-plant samples of the main cultivated variety"Zhanjiangchuanxinlian"of A.paniculata between the two cultivation modes.Furthermore,screening of marker metabolites and KEGG pathway enrichment analysis of differential metabolites were carried out.[Result](1)Most agronomic traits of the GAP group were superior to those of the WS group.The dry plant weight,fresh plant weight,and leaf area in the GAP group reached approximately twice of those in the WS group.In contrast,the WS group exhibited better performance in leaf drying rate and whole-plant drying rate(19.83%and 3.56%higher,respectively)as well as leaf ratio(9.54%higher).(2)The total andrographolide content of the GAP group was significant-ly higher,with increases of 27.90%,51.67%,and 40.76%in stems,leaves and whole plants,respectively,compared with that in the WS group.However,the WS group showed higher content of two specific andrographolide components,neoandrographolide and 14-deoxyandrographolide,with respective increases of 39.53%and 78.57%in whole plants.Both groups showed a consistent pattern that the total andrographolide content in leaves was higher than that in stems.The leaf ratios(32.39%and 35.48%)and total an-drographolide content(5.18%and 3.68%)of the two groups both met the standards specified in the Chinese Pharmacopoeia for me-dicinal materials(≥30%for leaf ratio and≥1.5%for total andrographolides)and decoction pieces(≥25%for leaf ratio and≥1.2%for total andrographolides).(3)Non-targeted metabolomics analysis identified a total of 5 566 metabolites in the dried whole-plant samples of the two groups,from which 2 602 differential metabolites were screened out.Among these metabolites,1 005 were up-regulated in the WS group and 1 597 were up-regulated in the GAP group.A total of 117 high-abundance marker differential me-tabolites were identified,including 37 belonging to 11 categories in the WS group and 80 belonging to 26 categories in the GAP group.KEGG pathway enrichment analysis indicated that these differential metabolites were significantly enriched in 45 metabolic pathways,including amino acid,nucleotide,carbohydrate,and lipid metabolism pathways.[Conclusion]A.paniculata under GAP cultivation exhibited significant advantages in yield and total andrographolide content,whereas that under WS cultivation showed dis-tinct characteristics in drying rate,leaf ratio,and content of some andrographolide components.The marker differential metabolites differed between the two cultivation modes,indicating potential for the development of products with different effects.WS cultivation could serve as an effective supplement to GAP cultivation.关键词
