光强对铁皮石斛多糖含量及相关差异表达基因的影响
Effects of different light intensities on polysaccharide content and related differentially expressed genes of Dendrobium officinale

Abstract
[Purpose]This study compared the effects of different light intensities on the polysaccharide content of Dendrobium offi-cinale and mined the related differentially expressed genes,aiming to provide reference for deciphering the molecular regulatory mechanism of polysaccharide synthesis in D.officinale.[Method]D.officinale was exposed to light at three intensities of 1 000 lx(weak light,CK),2 000 lx(moderate light),and 4000 lx(strong light),and growth and polysaccharide content were measured.Total RNA was extracted from the plant samples in the 1 000 lx and 4 000 lx groups.Transcriptome sequencing was performed and the differentially expressed genes(DEGs)were mined.Fluorescence qPCR was employed to validate the results.[Result]Different light intensities significantly affected growth and polysaccharide content of D.officinale,with the strong light group having the highest polysaccharide content.Under different light intensities,D.officinale produced a total of 1 501DEGs,which were mainly enriched in the starch and sucrose metabolism and the phenylpropanoid biosynthesis pathways.Among the 20 DEGs in the starch and sucrose metabolism pathways,3 up-regulated genes encoded β-glucosidase-22 and trehalose phosphate phosphatase,while 17 down-regulated genes encoded endoglucanase-like enzymes,pectinesterase-like enzymes,etc.Among the 332 promoter elements,176 were involved in the light response.[Conclusion]An increase in light intensity within the range of 1 000-4 000 lx can enhance the shoot fresh weight and polysaccharide content of D.officinale,and the genes involved in the starch and sucrose metabolism pathway play impor-tant roles in the regulation of polysaccharide accumulation in response to light.关键词
