ITGβ8在胃癌恶性进展中的临床价值:基于生物信息学与实验研究
Clinical value of ITGβ8 in the malignant progression of gastric cancer:Based on bioinformatics and experimental study

Abstract
Objective:To investigate the expression and clinical value of integrin β8(ITGβ8)in gastric cancer using bioinfor-matics databases and cellular experiments.Methods:The expression of ITGβ8 across various cancers was retrieved from the TIM-ER2.0 and UALCAN databases.The GEO,GEPIA,and HPA datasets were used to analyze ITGβ8 expression levels in gastric cancer.The Kaplan-Meier Plotter database was utilized to predict the prognosis of gastric cancer patients in relation to ITGβ8 ex-pression.The UALCAN database was used to examine the association between ITGβ8 expression and tumor stage in gastric can-cer patients.Bioinformatics enrichment datasets were employed to identify the biological signaling pathways of ITGβ8 in gastric cancer.ITGβ8 expression in gastric cancer tissues was assessed using immunohistochemistry(IHC)and quantitative reverse tran-scription PCR(qRT-PCR).The effects of ITGβ8 on gastric cancer cell phenotypes were investigated through siRNA transfection experiments,and epithelial-mesenchymal transition protein expression was analyzed by Western blot.Results:Bioinformatics da-tabases indicated that ITGβ8 was highly expressed across various cancers and significantly upregulated in gastric cancer(P<0.01).The Kaplan-Meier Plotter database revealed that higher ITGβ8 expression was associated with a poorer prognosis in gastric cancer patients.The UALCAN database demonstrated that ITGβ8 expression was significantly correlated with tumor stage and metasta-sis in gastric cancer patients(P<0.05).Bioinformatics enrichment analysis showed that ITGβ8 was involved in multiple biological processes and signaling pathways in gastric cancer progression(P<0.05).IHC and qRT-PCR data confirmed a higher ITGβ8 ex-pression in gastric cancer tissues(P<0.01).Compared to the control group,CCK-8,cell scratch,and Transwell assays indicated that the proliferation,migration,and invasion of MKN-74 cells were significantly reduced the siRNA-ITGβ8 group(P<0.01),while flow cytometry revealed a signifiant increase of MKN-74 cell apoptosis in the siRNA-ITGβ8 group(P<0.001).Western blot results showed that E-cadherin protein expression was significantly upregulated(P<0.01),whereas N-cadherin expression was downregulated in the siRNA-ITGβ8 group(P<0.05).Conclusion:ITGβ8 is highly expressed in gastric cancer and is closely associated with the occurrence and progression of the disease,suggesting that ITGβ8 may serve as a biomarker for early diagnosis and prognosis of gastric cancer.关键词
