电热针靶向针刺对颞下颌关节骨关节炎模型大鼠TLR4/NF-κB信号通路的影响
Effects of electrothermal acupuncture targeted intervention on the TLR4/NF-κB signaling pathway in temporomandibular joint osteoarthritis model rats

Abstract
Objective To investigate the mechanism of electrothermal acupuncture targeted intervention in a rat model of temporomandibular joint osteoarthritis(TMJOA)based on the Toll-like receptor 4(TLR4)/nuclear fac-tor-KB(NF-κB)signaling pathway.Methods 50 male Wistar rats were randomly divided into 5 groups(n=10 per group):blank control group,model group,electrothermal acupuncture group,warm acupuncture group,and conven-tional acupuncture group.The TMJOA model was established by injecting sodium iodoacetate solution into the upper cavity of the temporomandibular joint.Each intervention group received the corresponding treatment.The head-sha-king threshold was measured using a Von Frey pain measurement device.Histopathological changes in the tem-poromandibular joint condylar cartilage were observed via hematoxylin and eosin(HE)staining.Serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)were detected using enzyme-linked immunosorbent assay(ELISA).The mRNA and protein expression levels of TLR4 and NF-κB p65 in the tem-poromandibular joint condylar cartilage were measured using real-time quantitative polymerase chain reaction(RT-PCR)and Western blot analysis.Results Compared with the blank control group,the head-shaking thresholds of TMJOA model rats were significantly lower(P<0.01).Compared with the model group,the serum levels of IL-1β,IL-6,and TNF-α in the electrothermal acupuncture group were significantly lower(P<0.01),while the warm acu-puncture and conventional acupuncture groups showed significantly reduced serum levels of IL-6 and TNF-α(P<0.05 or P<0.01).Moreover,the electrothermal acupuncture group exhibited significantly lower values for all indica-tors compared to the warm acupuncture and conventional acupuncture groups(P<0.05 or P<0.01).Compared with the model group,the relative mRNA and protein expression levels of NF-κB p65 and TLR4 in the temporomandibu-lar joint condylar cartilage were significantly lower in the electrothermal acupuncture,warm acupuncture,and con-ventional acupuncture groups(P<0.01),with the electrothermal acupuncture group showing significantly lower le-vels than the warm acupuncture and conventional acupuncture groups(P<0.05 or P<0.01).Conclusion Electro-thermal acupuncture targeted intervention can inhibit the overactivation of the TLR4/NF-κB signaling pathway in the temporomandibular joint condylar chondrocytes of TMJOA model rats,downregulate the expression of down-stream inflammatory factors(such as TNF-α,IL-1β,and IL-6),thereby reducing the sustained stimulation and damage to chondrocytes caused by these inflammatory factors and delaying the progression of TMJOA.关键词
