脂多糖诱导脓毒症模型幼龄大鼠多脏器损伤评价研究
Evaluation of multi-organ injury in a lipopolysaccharide-induced juvenile rat sepsis model

Abstract
Objective To investigate the pathological alterations in the heart,liver,spleen,kidney,and brain tissues of juvenile rats with sepsis induced by lipopolysaccharide(LPS).Methods 24 4-6-week-old SPF-grade male Wistar rats were randomly divided into a blank group(intraperitoneal injection of normal saline)and a sepsis group(intraperitoneal injection of LPS 10 mg/kg),with 12 rats in each group.Three rats were sacrificed on the 1st,3rd,5th and 7th days after modeling,and the heart,liver,spleen,kidney and brain tissues were collected.Hematoxy-lin-eosin(HE)staining was used to observe the pathological damage,and the mRNA and protein expression levels of inflammatory factors signal transducer and activator of transcription 3(STAT3),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were detected by reverse transcription polymerase chain reaction(RT-PCR)and Western blotting(WB),respectively.Results After modeling,the various organs of juvenile rats exhibit characteristic patho-logical alterations:the heart tissue showed widened myocardial fiber spaces and inflammatory infiltration on the 1st day,the liver tissue showed progressive hepatocyte edema,the kidney tissue showed glomerular atrophy and brush border loss of renal tubules on the 1 st day,and the brain and spleen tissues showed neuronal degeneration and splenic follicular structure disorder.STAT3,TNF-α,and IL-6 showed organ-specific and time-dependent changes in mRNA and protein levels:STAT3 mRNA in the heart increased early but decreased later,and TNF-α protein was continuously highly expressed;IL-6 mRNA and protein in the liver continuously increased;TNF-α mRNA and pro-tein in the kidney were highly expressed throughout the process,and STAT3 protein was significantly decreased;TNF-α and IL-6 mRNA in the brain increased on the 7th day,and IL-6 protein was continuously highly expressed;STAT3 protein in the spleen continuously decreased,and IL-6 protein showed a trend of"early decrease-late in-crease".Conclusion This LPS-induced juvenile rat sepsis model successfully recapitulates the dynamic multior-gan pathology and inflammatory cytokine profiles seen in pediatric sepsis,offering a robust platform for mechanistic studies and therapeutic target exploration.关键词
