基于数据挖掘和网络药理学探讨中药治疗类风湿关节炎肝肾亏虚证的用药规律及作用机制
Investigation on prescription patterns and mechanisms of traditional Chinese medicine for rheumatoid arthritis with liver-kidney deficiency syndrome based on data mining and network pharmacology

Abstract
Objective To investigate the medication patterns of traditional Chinese medicine(TCM)in treating rheumatoid arthritis(RA)with liver-kidney deficiency syndrome and predict its potential therapeutic mechanisms.Methods Literature on TCM for RA with liver-kidney deficiency syndrome was retrieved from CNKI,VIP,and Wanfang databases.The TCM Inheritance Support Platform V2.5 was employed to analyze prescribing patterns.Net-work pharmacology identified chemical component targets of core herbs and disease targets.Intersecting genes be-tween core herbs and RA were subjected to Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses.Molecular docking validated interactions.Results Data mining identified 59 prescriptions and 146 herbs,characterized by warm,neutral,and cold natures;sweet,bitter,and pungent flavors;and primary meridian tropism.Association rule analysis identified Rehmanniae Radix Praeparata,Achyranthis Bi-dentatae Radix,Astragali Radix,Eucommiae Cortex,Angelicae Sinensis Radix,and Glycyrrhizae Radix as core herbs.Cluster analysis yielded 18 herb combinations.Network pharmacology revealed key components(sitosterol,stigmas-terol,β-sitosterol)and core targets(MAPK1,MAPK3,STAT3).GO analysis identified 1030 functional terms,while KEGG highlighted 174 signaling pathways(e.g.,PI3K/Akt,TNF).Molecular docking confirmed strong binding af-finity between key components and targets.Conclusion The therapeutic mechanism of core herbs for RA with liver-kidney deficiency syndrome may involve suppression of MAPK1,MAPK3 expression,regulation of immune-in-flammatory responses,and modulation of PI3K/Akt and TNF pathways.关键词
