Abstract
In recent years,infections caused by Photobacterium damselae subsp.damselae in aquatic animals have been increasing.Reliance on antibiotic treatment leads to drug resistance and food safety concerns.The combina-tion of inactivated vaccines with adjuvants can significantly enhance immune efficacy.This study aimed to evaluate the enhancing effects of three adjuvants(Astragalus polysaccharides,Freund's complete adjuvant,and 4%alumi-num hydroxide gel)on an inactivated P.damselae subsp.damselae vaccine.The optimal formaldehyde inactivation condition was first determined to be 0.4%(v/v)for 8 h.Zebrafish(Danio rerio)were then immunized via intrap-eritoneal injection and challenged.Results showed that the relative percent survival(RPS)of the inactivated vac-cine alone was 58.3%,while RPS values for vaccine groups combined with Astragalus polysaccharides,Freund's complete adjuvant,and aluminum hydroxide gel were 74%,45.8%,and 62.5%,respectively.Further monitoring of dynamic changes in non-specific immune indicators(ACP,AKP,CAT,SOD,MDA,LZM)revealed that the Astragalus polysaccharide-adjuvanted group significantly(P<0.05)and sustainably enhanced the host's immune response with good safety.In conclusion,Astragalus polysaccharide is the optimal adjuvant for enhancing the im-mune efficacy of the inactivated P.damselae subsp.damselae vaccine,providing an experimental basis for develo-ping efficient and safe aquaculture vaccines.关键词
