Effect of EMS Mutagenesis on Seed Germination and Seedling Phenotypic Characteristics of Oilseed Rape

Abstract
To create intermediate oilseed rape materials and investigate the effects of ethyl methanesulfonate(EMS)mutagen-esis on seed germination while determining the optimal mutagenesis treatment combination,seeds of Yunyouza No.15 were used as experimental material.Twelve treatment combinations were applied,including five EMS buffer solution concentra-tions(0.20%,0.40%,0.60%,0.80%,and 1.00%)and two exposure durations(2 h and 4 h).The results showed that as the EMS concentration increased,seed germination and seedling vigor of Yunyouza No.15 were significantly inhibited,with lon-ger exposure times amplifying this inhibitory effect.The concentration of 1.00%EMS with a 4 h exposure was identified as the half-lethal dose,representing the optimal mutagenesis treatment combination for oilseed rape seeds.Phenotypic evalua-tion of seedlings revealed variant phenotypes such as dwarfing,slender seedlings,leaf yellowing,purple or brown necrotic spots on leaves,and cotyledon malformation.关键词
