布莱克韦尔虫草的药物化学基础与抗乳腺癌潜力研究
Pharmacochemical Analysis and Potential Roles of Cordyceps blackwelliae on Anti-Breast Cancer

Abstract
For in-depth understanding of the bioactive components and pharmacological properties of Cordyceps blackwelliae,advanced methods by using the ultra-high-performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)in combination with the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)were employed in this study.Preliminary screening via the TCMSP database identified a total of 144 potential bioactive components,in which eleven characteristic components were finally confirmed to contain significant pharmacological activities on the basis of both oral bioavailability(OB≥30%)and drug-likeness(DL≥0.18).Association analysis of targets and diseases revealed a strong correlation between the C.blackwelliae's bioactive components and the human breast cancer,leading to identification of 45 intersection targets in total.Enrichment analysis of KEGG pathway demonstrated that genes of these targets were significantly enriched in a multiple number of pathways showing relations to cancers,including the apoptosis regulation pathway,IL-17-mediated inflammatory signaling pathway,and the PI3K-Akt-mediated cell proliferation and survival signaling pathway.In vitro experiments confirmed that the treatment of C.blackwelliae remarkedly altered the morphology of triple-negative breast cancer MDA-MB-231 cells and inhibited the cell viability,migration and invasion capabilities as well.This study elucidated the multi-component,multi-target and multi-pathway synergistic mechanism of C.blackwelliae against breast cancer that should provide a theoretical and practical basis for future development of novel and natural anti-breast cancer drugs.关键词
