Abstract

In order to fully exploit and utilize macrofungal resources,this study aims to investigate the feasibility of under-forest cultivation of Ganoderma in the northern Shaanxi area of China.A wild Ganoderma strain,collected from Mohe of Heilongjiang Province,was isolated and purified.Morphological characterization and molecular biological analysis identified this strain as belonging to the species of Ganoderma tsugae.Effects of the different carbon sources,Tsuga sawdust extract concentration,pH value and temperature of culture medium on its mycelial growth of G.tsugae were then examined.Single-factor experiments revealed that the optimal conditions of cultivation for mycelial growth of mother-strain were to use glucose as the carbon source,a concentration of 60 g/L of Tsuga sawdust extract,a temperature of 25℃ and a value of pH 5.0.Selection of three optimal levels to optimize above factors by using the response surface methodology(RSM),furthermore,suggested the ideal conditions for mycelia growth of G.tsugae that included a concentration of 61 g/L Tsuga sawdust extract,26℃,and pH 5.0.Under these conditions,the mycelia morphologically grew dense and revealed a growth rate reaching to 8.15 mm/d,more than two-fold higher than that of the control group.Comparative analysis revealed a lower yield but significantly higher bioactive compound contents of G.tsugae fruitbody cultivated under the natural forests than that within the greenhouse,in which the content of triterpenes and polysaccharides detected from the samples grown under-forest was 49.94%and 50.91%higher than that grown within greenhouse,respectively.These findings should provide a scientific basis for optimizing culture conditions of mother-strain and under-forest cultivation of G.tsugae that,furthermore,give valuable references for the future research and development of other Ganoderma species.
