Screening of Culture Media Suitable for Fresh Consumption of Cordyceps militaris by Sensory Evaluation and Metabolite Analysis

Abstract
This study aimed to screen out Cordyceps militaris suitable for fresh consumption and sales by using culture media made from rice and wheat.Two purchased strains of Cordyceps militaris,the southern rice substrate C.militaris strain(CmSF)and the northern wheat substrate C.militaris(CmNF),were subjected to the analysis of their genetic relationship using ITS sequences,and to the examination of taste,appearance and texture carried out by 200 evaluators under three conditions:eating fresh,boiling for 5 minutes and home-cooking.In combination with targeted and non-targeted metabolomic approaches,moreover,the differences of metabolites between above two strains of C.militaris grown under two different kinds of culture media were also analyzed.Analysis of the ITS sequences demonstrated a very close genetic relationship between the two strains,while the sensory evaluation results showed that CmNF was more favored than CmSF.Conduction of targeted and non-targeted metabolomic analysis in the fruiting bodies of CmNF and CmSF led to successful screening of a total number of 45 differential metabolites,which contained 30 organic acids and their derivatives(including seven specific to CmNF and three specific to CmSF),five monosaccharides,two disaccharides,two organic compounds,three nucleotides,one fatty acid,and one of each lipid,lipoid,and ester substances.In comparison with CmSF,CmNF strain revealed nine up-regulated and 26 down-regulated metabolites.Based on our results,it was concluded that the main reasons why CmNF was better suited for eating fresh were owing to its superior appearance and taste as well as its enhanced preservation ability of freshness.Our research thereafter provides a useful foundation for future development of industrial cultivation and food processing of C.militaris,and a theoretical framework to support future scientific analysis of mechanisms for the effects of different culture substrates on differential metabolites of C.militaris as well.关键词
