Analysis of Morphological Features and Esterase Isozymes of Inonotus hispidus Fruting Body at Different Growth Stages

Abstract
In order to provide the scientific basis for utilization of genetic resources and development of quality standards of medicinal fungi,morphological and pharmacognostic features of Inonotus hispidus fruiting bodies collected respectively at the budding,maturing and aging stages were characterized and compared systematically in this study through the macro and microscopical investigations as well as the detection of thin layer chromatography and esterase(EST)isozyme activity.Our results showed that most of the hyphae at the budding stage had a light brown or dark brown color with thin cell wall in which some curved hyphae seemed to carry the yellowish brown substances.At the maturing and aging stages,on the other hand,the hymenium of hyphae revealed a deepen dark brown color with the thickest cell wall.At the aging stage,furthermore,there existed curved and rope-shaped hyphae intertwining with each other.Based on the control of hispidin standard,TLC analysis detected the same fluorescent spots from I.hispidus methanol extracts at the corresponding positions between the budding and maturing stages which,however,could not be seen for the aging stage.Examination of EST isozyme activity of I.hispidus indicated clearly its specificity to the different developmental stages,including the numbers and denseness of isozyme bands.No significant EST isozyme bands were observed at the hyphae stage.The maximum number of EST isozyme bands were detected at the aging stage with five bands whose color was much deeper than those observed at the budding and maturing stages,reflecting that the activity of EST isozymes was facilitated at the aging stage.关键词
