骨肉瘤诱导多能干细胞模型的制备
Induced pluripotent stem cell model of osteosarcoma

Abstract
Objective To explore the application of induced pluripotent stem cell(iPSC)technology in osteosar-coma stem cells(OSC),the construction of osteosarcoma-induced pluripotent stem cells(OS iPSC)and the character-ization their properties are of great significance for accelerating innovation and developing tailored intervention strate-gies for osteosarcoma.Methods Lentiviral particles were used to infect mature osteosarcoma cells HOS to construct OS iPSC.Alkaline phosphatase(ALP)staining and immunofluorescence staining were performed on OS iPSC to iden-tify its pluripotency.Suspension culture of OS iPSC was used to prepare embryoid bodies(EB)to identify its in vitro differentiation ability.OS iPSC was directionally differentiated into mesenchymal stem cells(MSC),followed by evaluation of OS iPSC-derived MSCs(OS iPSC-MSC)using immunofluorescence staining.OS iPSC-MSC was induced to differentiate into osteoblasts to simulate the pathogenesis of osteosarcoma.ALP and alizarin red staining(ARS)were used to observe the osteogenic differentiation process.Results OS iPSC was generated after high-titer lentiviral par-ticles(7.8×106 IU/µL)infected osteosarcoma cells.The cells showed strong positive ALP and could maintain an un-differentiated state in mTeSR1 medium.Immunofluorescence staining results showed that OS iPSC expressed pluripo-tency factors OCT4,NANOG and surface antigen SSEA4.After 7 days of suspension culture of OS iPSC,EBs could be formed.After directional induction culture of OS iPSC,mature OS iPSC-MSC could be differentiated after 43 days.Immunofluorescence staining results showed that the surface markers CD44,CD73,CD90 and CD105 of OS iPSC-MSC were positive.On the 9th day of osteogenic induction and differentiation of OS iPSC-MSC,ALP and ARS were positive.And as the differentiation time progressed,more and more ALP and mineral deposits could simulate the pathogenesis of osteosarcoma.Conclusion In this study,HOS is successfully reprogrammed to construct an OS iPSC model,which can be used as an in vitro cell model for studying the pathogenesis of osteosarcoma and is expected to pro-vide a theoretical basis and experimental model for studying the occurrence and development of osteosarcoma and de-veloping targeted drugs.关键词
