摘要

Abstract

Objective To determine the mechanism of Xuefuzhuyu Decoction in treatment of retinal vein occlusion(RVO)by using network pharmacology,and to verify it by molecular docking.Methods Multiple public databases were utilized to find and screen Xuefuzhuyu Decoction active ingredient targets and RVO disease targets,whose common targets were then used as potential therapeutic targets.The network of"tradi-tional Chinese medicine-active ingredient-potential target"was established and analyzed via by topological parameters to screen the main active ingredients of Xuefuzhuyu Decoction.The protein interactions network of common targets of drugs and diseases was constructed,and the core targets identified.The gene ontology function and Kyoto Encyclopedia of Genes and Genomes enrichment of the common targets were analyzed,and molecular docking of the main active ingredients and core targets finally carried out.Results A total of 157 active ingredients were identified in Xuefuzhuyu Decoction,and 144 potential targets of Xuefuzhuyu Decoction for the treatment of RVO determined,and the main enriched pathways included in the interleu-kin-17 and the tumor necrosis factor signaling pathway,etc.The key molecules include quercetin,luteolin,kaempferol,wogonin and naringenin,etc.signal transducers and activators of transcription 3,mitogen-acti-vated protein kinase 3,transcription factor Jun,mitogen-activated protein kinases 1,and tumor protein 53 were the core therapeutic targets of the soup.The five core targets had good binding activities with the main active ingredients.Conclusion Xuefuzhuyu Decoction may act on key targets such as signal transducers and activators of transcription 3 through quercetin and lignans,etc.,and alleviate the adverse effects of oxidative stress injury,endothelial cell injury,atherosclerosis and lipid deposition through the modulation of signaling pathways,such as interleukin-17,so as to treat RVO.
