Causal relationship between psychiatric disorders and chronic kidney disease:A Bidirectional Two-Sample Mendelian Randomization analysis

Abstract
To inveatigate the causal relationship between five psychiatric disorders and chronic kidney disease(CKD),this study employed a bidirectional two-sample Mendelian randomization(MR)analysis.Instrumen-tal variables(IVs)were selected using a threshold of P<1.0×10-5,and analyses were conducted using inverse-variance weighted(IVW),MR-Egger,weighted median,simple mode,and weighted mode meth-ods.This study explored causal links between CKD and five psychiatric disorders:major depressive disorder,anxiety disorder,post-traumatic stress disorder(PTSD),cannabis disorder,and schizophrenia.A significant causal association was identified only between PTSD and CKD(OR=1.07,95%CI:1.02～1.13,P=0.009),with no significant causal relationships observed in other directions.These findings remained consis-tent across different IV selection thresholds,and sensitivity analyses confirmed their robustness.This study establishes PTSD as a direct causal risk factor for CKD,while demonstrating no significant causal asso-coations between CKD and major depressive disorder,ansiety disorder,cannabis use disorder,or schizophre-nis.These findings provide a scientific basis for CKD monitoring in PTSD patients,emphasizing the need for integrated psychiatric and renal management in clinical practice.Furthermore,this work clarifies precise inter-vention targets,helping to avoid unnecessary medical interventions for comorbid conditions without causal evidence.关键词
