Abstract
This experiment was designed to investigate the effects of probiotics,prebiotics on the growth per-formance,intestinal epithelial cell differentiation and intestinal health of broiler chickens.A total of 180 one-day-old male broilers with an average body weight of(43.56±0.11)g were randomly divided into three groups with six replicates per group and 10 chickens per replicate.The trial was divided into three stages,the first stage is 1-10 d,the second stage is 11-21 d,and the third stage is 22-42 d.In the first stage of the experiment,all broilers in the three groups were fed with a corn-soybean basal diet.In the second and third stages of the experiment,the broilers in the control group were fed with corn-soybean basal diet,and the broiler chickens in the two treatments were fed the basal diet supplemented with 300 mg/kg probiotics(Bacillus subtilis,viable count 3×108 CFU/kg)or 700 mg/kg prebiotic(6%mannose oli-gosaccharides and 8%β-glucan)respectively.The results showed that broilers in the probiotics group had higher body weight(at 21 d and 42 d),average daily feed intake and average daily gain(11-21 d and 11-42 d)compared with broilers in the control group(P<0.05);broilers in the prebiotics group had higher body weight at 42 d and average daily gain during 11-42 d compared with broilers in the control group(P<0.05).Probiotics and prebiotics significantly increased the jejunal villus height,jejunal villus height to crypt depth ratio and the mRNA expression of jejunal occludin(Occludin),claudin-2(Claudin-2)and claudin-3(Clau-din-3)in the jejunum of broilers(P<0.05).Probiotics and prebiotics significantly increase the mRNA expression of homeodomain protein(Hopx),epithelial cell adhesion molecule(EPCAM),chromogranin A(Chga)and sucrase isomaltase(SI)in the jejunum of broilers(P<0.05).The mRNA expression of goblet cells differentiation regulating factor Kruppel-like factor 4(Klf4)in the probiotics and prebiotics groups were sig-nificantly up-regulated in the jejunum of broilers(P<0.05).The mRNA expression of proton coupled oligo-peptide transporter 1(PepT1),solute carrier family(SLC6A19,SLC36A1 and SLC38A4)in the probiotics and prebiotics groups were significantly increased in the jejunum of broilers(P<0.05).The mRNA expression of toll-like receptor 2(TLR-2),interleukin 1β(IL-1β)and interferon γ(INF-γ)in the probiotics and prebiotics groups were significantly down-regulated in the jejunum of broilers(P<0.05).Probiotics and prebiotics had no significant effect on the relative mRNA expression levels of apoptosis-related genes,including cysteinyl aspartate specific proteinase-1(caspase-1),Bcl2-associated X protein(Bax),and B-cell lymphoma-2(Bcl-2),in the jejunum of broilers(P>0.05).In conclusion,dietary addition of probiotics or prebiotics effectively promoted the differentiation of jejunal epithelial cell,enhanced the secretion and absorption function of the jejunal epithelial cell,and decreased the level of jejunal inflammation,leading to the improvement of growth performance in broilers.关键词
