长期接种根瘤菌替代氮肥对东北黑土团聚体细菌群落及胞外酶活性的影响
Effects of long-term inoculation of rhizobium instead of nitrogen fertilizer on bacterial community and extracellular enzyme activity in aggregates of black soil in northeast China

Abstract
In order to explore the influence mechanism and coupling relationship of long-term inoculation of rhizobia instead of nitrogen fertilizer on the bacterial community and extracellular enzyme activity in the soil aggregate micro-regions,the long-term black soil location test field of Jilin Academy of Agricultural Sciences was taken as the research object.Four soil samples were collected,including no fertilization(CK),phosphorus and potassium fertilization(PK),nitrogen,phosphorus and potassium fertilization(PK+N),phosphorus and potassium fertilization and inoculation of Bradyrhizobium japonicum 5821(PK+R).The effects of different fertilization treatments on the stability,chemical properties,bacterial community structure,and extracellular enzyme activity of different particle size aggregates in northeastern black soil were analyzed.The results indicated that fertilization altered the distribution characteristics of soil aggregates,reducing their stability and hindering the restoration of black soil quality.The PK+N and PK+R treatments significantly increased the total nitrogen content of aggregates and the activities of β-1,4-N-acetylglucosaminidase and β-D-cellobiohydrolase,thereby improving the carbon and nitrogen cycling in the black soil.The application of chemical fertilizers combined with rhizobia inoculation significantly enhanced the alpha diversity index of bacterial communities and markedly altered their structure,increased the relative abundance of Bradyrhizobium and Sphingomonas genera,which was beneficial for promoting the decomposition and transformation of organic matter in black soil.The bacterial community composition of aggregates with different particle sizes was basically consistent.Redundancy analysis and Mantel test analysis showed that acid phosphatase activity,β-D-cellobiohydrolase activity,and total soil carbon content were the main environmental factors affecting bacterial communities.In summary,the application of chemical fertilizers increased the carbon and nitrogen contents and extracellular enzyme activity of black soil,but it reduced the stability of soil aggregates and affected the bacterial community.Inoculating rhizobium instead of nitrogen fertilizers could reduce the decrease of aggregate stability to some extent,effectively increase the total nitrogen content of soil and improve the structure of bacterial community.关键词
