Abstract

To explore the function of Trehalose-6-phosphate phosphatase(TPP)in yam growth and the expression level changes of its regulatory genes under abiotic stress conditions,this study used'Dahechangyu'and'Bikeqi'yams as experimental materials,combined with transcriptome data to clone the yam TPP6 gene.The gene underwent bioinformatics analysis and subcellular localization,fol-lowed by overexpression in tobacco through genetic transformation.To assess the gene's function,transgenic tobacco was subjected to abiotic stress conditions followed by analysis using qPCR and oth-er methods.The results showed that DoTPP6 contained a complete open reading frame of 1 152 bp,encoding 383 amino acids as an unstable hydrophilic protein;subcellular localization showed that the gene was located in the nucleus and plasma membrane.The DoTPP6 gene from yam shared 96.87%sequence similarity with the homologous gene in winged yam,indicating a close phylogenetic relation-ship.DoTPP6 expression was highest in the tubers after 165 days of cultivation,which was conducive to starch accumulation.Under low temperature and drought stress treatments,the expression levels of DoTPP6 in yam-cultured seedlings increased by 0.51 times and 0.47 times,respectively,while in DoTPP6-transgenic tobacco plants,it increased by 6.96 times and 0.99 times,respectively,compared to the control.These results indicate that DoTPP6 plays a role in abiotic stress response and could serve as a candidate gene for improving yam stress resistance.
