化痰通络法对咳嗽变异性哮喘豚鼠肺组织血管重构的影响
Effect of therapy for resolving phlegm and unblocking collaterals on vascular remodelling in guinea pigs with cough variant asthma

Abstract
Objective It is to study the effect of therapy for resolving phlegm and unblocking collaterals on vascular re-modelling in lung tissue of guinea pigs with cough variant asthma(CVA)based on vascular endothelial growth factor(VEGF)/Notch signaling pathway,and to explore the mechanism of this therapy in the treatment of CVA.Methods Forty guinea pigs were randomly divided into normal group,model group,Montelukast sodium group and resolving phlegm and unblocking collaterals group,with 10 animals in each group.The guinea pigs in all groups except for the normal group were sensitized with ovalbumin and aluminum hydroxide and stimulated with ovalbumin inhalation to replicate CVA models.At 30 minutes before each stimulation,the Montelukast sodium group was given Montelukast sodium suspension at a dose of 0.1 mg/mL by gavage,the resolving phlegm and unblocking collaterals group was given compound solution for resolving phlegm and unblocking collaterals at a dose of 0.74 g/mL by gavage,the control group and model group were given drink-ing water by gavage,all administered once daily,continuously treated for 14 days.The vascular pathological morphology of lung tissue was observed by HE staining,the total vascular area,lumen area,vascular smooth muscle layer area,vascular outer diameter,and vascular smooth muscle thickness in lung tissue vessels were detected,the ratios of lumen area/total vascular area,vascular smooth muscle layer area/total vascular area,vascular smooth muscle thickness/vascular outer di-ameter were calculated,the protein and mRNA expressions of hypoxia-inducible factor-1α(HIF-1α),VEGF,DLL4,Notch4 in lung tissue were respectively detected by Western blot and real-time quantitative PCR.Results Compared with the normal group,the vascular wall of lung tissue was thickened and the hyperplasia of smooth muscle of media layer was obvious in the model group;there were no obvious changes in the total vascular area and vascular outer diameter in lung tis-sue(all P>0.05),however,the lumen area and lumen area/total vascular area were significantly decreased(all P<0.05),the vascular smooth muscle layer area,vascular smooth muscle layer area/total vascular area,vascular smooth muscle thickness,and vascular smooth muscle thickness/vascular outer diameter were all significantly increased(all P<0.05);the relative protein and mRNA expressions of HIF-1α,VEGF,and DLL4 in lung tissue were significantly increased(all P<0.05),while the relative protein and mRNA expressions of Notch4 were significantly reduced(all P<0.05).Compared with the model group,the vascular wall and smooth muscle of media layer in lung tissue were thinner in the Mon-telukast sodium group and resolving phlegm and unblocking collaterals group;there were no obvious changes in the total vascular area and vascular outer diameter in lung tissue(all P>0.05).Compared with the model group,the lumen area and lumen area/total vascular area in the resolving phlegm and unblocking collaterals group were significantly increased(all P<0.05),the vascular smooth muscle layer area,vascular smooth muscle layer area/total vascular area,vascular smooth muscle thickness,and vascular smooth muscle thickness/vascular outer diameter were all significantly decreased(all P<0.05);the relative protein and mRNA expressions of HIF-1α,VEGF,and DLL4 in lung tissue were significantly de-creased(all P<0.05),while the relative protein and mRNA expressions of Notch4 were significantly increased(all P<0.05).Conclusion Therapy for resolving phlegm and unblocking collaterals can inhibit the proliferation of vascular smooth muscle cells,improve vascular remodeling,thus to reduce airway remodeling in guinea pigs with cough variant asthma pos-sibly via down-regulating the expressions of HIF-1α,VEGF and DLL4 in lung tissue and up-regulating the expression of Notch4.关键词
