养血通脉方对利血平诱导低血压大鼠氧化应激及循环肾素-血管紧张素系统稳态的影响
Effects of decoction for nourishing blood and unblocking pulse on oxidatiue stress and RAS homeostasis equilibrium in rats with hypotension induced by reserpine

Abstract
Objective It is to explore the effects of decoction for nourishing blood and unblocking pulse on oxidatiue stress and circulatory renin-angiotensin system(RAS)homeostasis in rats with hypotension induced by reserpine.Methods One hundred male Wistar rats were taken,in which 10 rats were randomly selected as the blank control group,and the re-maining 90 rats were gavaged with rifampicin suspension 0.032 mg/kg twice a day until the end of the experiment.The 50 rats that successfully established hypotension model by continuous gavage of rifampicin for 3 weeks were randomly divided into model control group,midodrine hydrochloride group and low,medium and high dose groups of decoction for nourishing blood and unblocking pulse,with 10 rats in each group.The midodrine hydrochloride group was given midodrine hydrochlo-ride solution 0.45 mg/(kg·d)by gavage,the low,medium and high dose groups of decoction for nourishing blood and unblocking pulse were given decoction for nourishing blood and unblocking pulse of respective dose by gavage,while the blank control group and model control group were given pure water 10 mL/(kg·d)by gavage.The changes in body weight,systolic blood pressure(SBP)and heart rate of the rats during the intervention period were observed,the levels of superoxide dismutase(SOD),malondialdehyde(MDA)in serum and brain tissues,and the levels of renin,angiotensin-converting enzyme(ACE),and angiotensin Ⅱ(Ang Ⅱ)in serum of rats in each group were measured at the end of 4 weeks of intervention.Results Compared with the blank control group,the weight gain of the rats in the model control group was slow,the SBP was decreased,the heart rate was increased,the SOD levels in serum and brain tissue were signifi-cantly decreased(all P<0.05),the MDA levels in serum and brain tissue,and the levels of Renin,ACE,AngⅡin ser-um were significantly increased(all P<0.05).During the intervention period,the rats in the midodrine hydrochloride group and each decoction for nourishing blood and unblocking pulse group showed improved body weight gain,increased SBP,and decreased heart rate,with the greatest increase of SBP in the midodrine hydrochloride group in the first 2 weeks,followed by a smooth recovery of SBP in each decoction for nourishing blood and unblocking pulse group in the second 2 weeks,and a slight decrease in the midodrine hydrochloride group.Compared with the model control group at the end of 4 weeks of intervention,the body weight of rats in the middle and high dose groups of decoction for nourishing blood and un-blocking pulse and midodrine hydrochloride group,and the SBP of rats in each decoction for nourishing blood and unblock-ing pulse group were significantly increased(all P<0.05);the levels of serum SOD of rats in each treatment group,and the levels of SOD in brain tissues in the middle and high-dose groups of decoction for nourishing blood and unblocking pulse were significantly increased(all P<0.05),the levels of serum MDA,ACE and Ang Ⅱ of rats in each treatment group and the levels of MDA in brain tissues of rats in the middle and high dose groups of decoction for nourishing blood and un-blocking pulse and midodrine hydrochloride group were all significantly decreased(all P<0.05).Conclusion Decoction for nourishing blood and unblocking pulse can improve the body weight,blood pressure and heart rate of rifampicin-induced hypotensive rats,and its blood pressure increasing effect is relatively slower but smoother than that of midodrine hydrochlo-ride,meanwhile,it has good improvement effects in the alleviation of oxidative stress in the body and the regulation of cir-culating RAS homeostasis.关键词
