Mendelian Randomization Analysis of the Causal Relationship Between Blood Lipid Metabolism Level and Dementia Classification

Abstract
Objective To explore the causal relationship between lipid metabolism and dementia by two-sample Mendelian randomization(MR)method.Methods MR analysis was performed using inverse variance weighting(IVW),weighted median method,MR-Egger regression and weighted model.OR value and 95%CI were used to evaluate whether there was a correlation between common blood lipid indexes[total cholesterol(TC),triglyceride(TG),apolipoprotein A(ApoA),apolipoprotein B(ApoB),low-density lipoprotein(LDL),high-density lipoprotein(HDL)]and Alzheimer's disease dementia(AD)and vascular dementia(VAD).Results MR analysis showed that LDL had a positive causal relationship with AD,OR=1.2460(95%CI:1.0367-1.4977,P=0.0191).There was no correlation between other blood lipid indexes and AD and VAD(P>0.05).Conclusion Based on the analysis results,there is a positive causal relationship between LDL and AD.The higher the LDL level,the higher the risk of AD.There is no evidence to support the causal relationship between other blood lipid indexes and AD and VAD.关键词
