Abstract

Objective To investigate the inhibitory effect of the traditional Chinese medicine Leucas ciliata Benth.extract on lipopolysaccharide(LPS)-induced nitric oxide(NO)expression in RAW264.7 cells and its molecular mechanism.Methods The impact of different concentrations of Leucas ciliata Benth.extract on RAW264.7 cell viability was assessed using the CCK-8 assay to determine the safe and effective concentration range.The Griess method was used to measure NO release levels in cell culture supernatants,reflecting the regulatory effect of the Leucas ciliata Benth.extract on LPS-induced NO expression in RAW264.7 cells.Results At concentrations up to 200 μg/ml,Leucas ciliata Benth.extract showed no significant effect on RAW264.7 cell viability(P>0.05).Concentrations of 25 μg/ml and higher significantly inhibited NO release in LPS-stimulated RAW264.7 cells(P<0.001),exhibiting a concentration-dependent effect.Conclusion Leucas ciliata Benth.extract effectively inhibits LPS-induced excessive NO expression in RAW264.7 cells,with the inhibitory effect closely correlated with the concentration of the extract.
