红萸饮对三硝基苯磺酸诱导肠纤维化小鼠的干预作用及对TGF-β1/Notch1信号通路的影响
Therapeutic effect of Hongyu Yin on intestinal fibrosis induced by trinitrobenzene sulfonic acid in mice and its effect on TGF-β1/Notch1 signaling pathway

Abstract
Objective It is to observe the therapeutic effect of Hongyu Yin on trinitrobenzenesulfonic acid(TNBS)-in-duced intestinal fibrosis in mice,and to explore its possible mechanism based on transforming growth factor-β1(TGF-β1)/Notch1 signaling pathway.Methods Sixty C57BL/6 mice were randomly divided into 4 groups:control group,model group,Hongyu Yin low dose group and Hongyu Yin high dose group,with 15 mice in each group.The mice in the control group were not intervened,and the mice in the other three groups were induced by TNBS enema at weekly increasing con-centration gradient,the Hongyu Yin low dose group and Hongyu Yin high dose group were given Hongyu Yin at the dose of 11.875 g/(kg·d),23.75 g/(kg·d)by gavage every day,continuously treated for 6 weeks.The mice in each group were sacrificed on the 37th day of experiment,the serum levels of interleukin-13(IL-13)and interleukin-17A(IL-17A)were measured by ELISA,the intestinal fibrosis was assessed by HE staining and Masson's staining,the positive expres-sions of α-smooth muscle actin(α-SMA)and collagenⅠin colon tissue were detected by immunohistochemistry,the pro-tein expressions of TGF-β1 and Smad3 in colon tissue were detected by Westernblot,and the mRNA expressions of Notch1,Hes1,and Hey1 in colon tissue were detected by Real-time qPCR.Results Compared with the control group,the serum levels of IL-13 and IL-17 A in the mice of the model group were significantly increased(all P<0.05);there was a large number of inflammatory cell infiltration in colon tissue,and the collagen fibers in submucosa were significantly proliferated;the mean optical density values of positive expressions of α-SMA and Collagen Ⅰ in colon tissue,as well as the relative protein expressions of TGF-β1 and Smad3 and the relative mRNA expressions of Notch1,Hes1,and Hey1 were all signifi-cantly increased(all P<0.05).Compared with the model group,the serum levels of IL-13 and IL-17A were significantly reduced in the mice of the Hongyu Yin low dose group and Hongyu Yin high dose group(all P<0.05);the inflammatory cell infiltration and collagen fiber deposition in colon tissue were reduced;the mean optical density values of positive ex-pressions of α-SMA and CollagenⅠ,the relative expressions of TGF-β1 and Smad3,and the relative mRNA expressions of Notch1,Hes1,and Hey1 in colon tissue were all significantly decreased(all P<0.05).Furthermore,the colon tissue in-jury was the least severe,and the decreases of these indicators were the most obvious in the Hongyu Yin high dose group(all P<0.05).Conclusion Hongyu Yin can improve TNBS-induced intestinal fibrosis in mice,and its mechanism may be related to inhibiting the activation of Notch1 signaling pathway and down-regulating the expression of TGF-β1/Smad signa-ling pathway.关键词
